Objective:By establishing a model of ischemia-reperfusion injury in rats, observed in rats after ischemia-reperfusion injury of the liver structure, function and expression of inflammatory factors changes, and observation and discussion of apoptosis and Autophagy in liver ischemia-reperfusion play a role in the process. Study of Berberine on hepatic ischemia-reperfusion injury has a protective effect, and for clinical work for reducing hepatic ischemia-reperfusion injury after surgery to provide theoretical support and effective drug treatments.Methods:The experimental group:1. healthy, clean SD rats,54 weeks 8-12 weeks, weight 250-280g, regardless of male and female, were randomly divided into 3 groups. Sham-operated rats, respectively (SHAM), ischemia-reperfusion rat (HIRI), Berberine intervention in rats with ischemia reperfusion Group (HIRI+BER).18 rats in each group. SHAM rats only switch operation operation operation without liver ischemia, HIRI rat hepatic ischemia-reperfusion injury surgery, ischemic 60min,HIRI+BER rats for 14 days before surgery to Berberine (60 mg/kg) stomach, once a day, also ischemic-reperfusion surgery, ischemia time is 60min.Monitoring three sets of indexes of postoperative changes in SD rats, three groups of rats were divided into three separate time periods,1H,6H,12H three times, each time period is divided into 6 rats. Cutting rats with left lateral lobe of the liver and collected venous blood specimen, measurement of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) indicators. Detection of inflammatory cytokines HMGB1,IL-6,TNF-a by ELISA. Western blot analysis of Autophagy and apoptosis related protein in liver tissue Bax, Beclin-1, expressions such as Bcl-2,Caspase-3,Caspase-7, HE dyed the liver, microstructure changes of liver cells. Results:1.Wake up after surgery in rats, stand up, and maintain effective breathing and pulse showed that successful operation.2.Hepatic expression of apoptosis-related protein Bax, Caspase3,7, three time periods in rats:the SHAM group and HIRI+BER significantly lower than that of HIRI (P<0.05), HIRI+BER group than in the HIRI was significantly reduced (P<0.05). Inhibit the expression of apoptosis-related protein Bcl-2 in the liver, rats in three time periods:SHAM groups and HIRI+BER groups were significantly higher than that of HIRI (P<0.05), significantly higher than that of HIRI HIRI+BER Group (P<0.05). Autophagy-related protein markers in liver tissue ofBeclin-1 expression of rats in three time periods:the SHAM group and HIRI+BER significantly lower than that of HIRI (P<0.05), HIRI+BER group than in the HIRI was significantly reduced (P<0.05).3.Changes of serum ALT and AST levels:reactions on clinical indicators of serum ALT and AST are important indicators of liver function, three time periods in rats:comparison of SHAM group, HIRI group, and HIRI+BER of serum ALT and AST levels increased significantly (P<0.05), and indicators of serum transaminase after restore perfusion 6h peaked after the fall. HIRI group and HIRI+BER group, decreased serum ALT and AST levels (P<0.05).4 Inflammatory factors in serum of HMGB1, IL-6, TNF-a index, rats in three time periods: HIRI group and HIRI+BER group, HMGB1, IL-6, TNF-a index significantly higher than that of SHAM (P<0.05). Compared with HIRI, HMGB1, IL-6, HIRI+BER group, TNF-a index significantly reduced (P<0.05).5 HE staining of liver tissue:liver cells have normal SHAM group, the structure is clear, liver cell cord neatly. Reperfusion 1H in rats:HIRI mild swelling of rat liver cells, HIRI+BER group no significant damage in rat hepatocytes; reperfusion 6H in rats:HIRI severe edema in rat liver cells, liver disorder and cable structures, seen ballooning degeneration and necrosis, inflammatory cell infiltration in more. HIRI+BER edema in rat liver cells, local small amounts of liver cell degeneration and necrosis. Reperfusion 12H in rats:HIRI+BER rat liver edema and necrosis, level better than that of HIRI.Conclusion:1.The Berberine inhibits hepatic ischemia-reperfusion injury in Autophagy and apoptosis expression decreased the release of inflammatory cytokines in serum.2.1n hepatic ischemia-reperfusion injury in the Suppression of Autophagy and apoptosis in expression and release of inflammatory factors in blood serum reduction Improves liver function, plays a role in protecting the liver.3.The Berberine by inhibiting apoptosis, and Autophagy reducing hepatic ischemia-reperfusion injury in rats.
