Objective:To explore the possible active substances in different Eucalyptus leaves by studying the components and the anti Echinicoccus Granulosus protoscolices efficacy of the extracts from the Eucalyptus leaves, and establishing the chromatogram-activity relationship.Method:1.Harvesting mature leaves of Eucalyptus Robusta and Eucalyptus Grandis on the same date in each month in one year and then drying naturally;2.Ultrasonic extract the leaveswith 4 different polar solvent petroleum ether, dichloromethane, ethyl acetate and anhydrous ethanol; 3.Protoscolices were incubated with different extracts at various concentrations to test the efficacy and LC50 were calculated; 4. Analyzing the components of every extracts through gas chromatography-mass spectrometry (GCVMS).The content of the component was determindedsemiquantitatively by calculating the normalized peak area and relative peak area; 5. Partial least two regression analysis was applied to analyze the chromatogram-activity relationship. Rank the regression parameters of each ingredient and speculate on the chemical composition of the leaf extract which owned the most effect of anti-protoscolices activity.Result:1. There were different composition and content in these two species of Eucalyptus leaves. The species number in petroleum ether extract were roughly the same, but in dichloromethane extracts,the number of species in Eucalyptus robusta leaves was averaged 49 species, Eucalyptus grandis was 73. The number of peaks included in the statistical analysis inpetroleum ether extracts of Eucalyptus robusta leaveswas 33, in dichloromethane extract of Eucalyptus robusta leaf was 46, and in petroleum ether and dichloromethane extracts of Eucalyptus grandis leaves were both 32.2. In the inhibition of protoscolices activity test, the petroleum ether and dichloromethane fractions had obvious effect, but the effect of ethyl acetate and ethanol extracts were slight. Among the petroleum ether extracts of Eucalyptus robusta leaves, the sample from June showed best anti Echinicoccus granulosus effect. Its LC50 was 2.577 μg/ml. The sample from December among Eucalyptus grandis leaves petroleum ether extracts was best, and its LC50 was 3.110 μg/ml. The sample from November was the best one in Eucalyptus robusta dichloromrthane extractives against Echinicoccus granulosus effect, The LC50 was 21.85μg/ml. And in Eucalyptus r grandis dichloromethane leaves extracts, the sample from March was the best one, its LC50 was 2.679.g/ml.3.The results of chromatogram-activity relationship showd that, the parameters value of different components for the protoscolex mortality werenot identical, but some parameters values are very close to each other.Conclusion:The killing activity of protoscolex of eucalyptus leaf extractwas the combined effect of some components.
