Objective: To observe the protective effect of Ginkgo biloba extract on hepatic injury induced by intestinal ischemia-reperfusion in rats, and to explore the impact of Ginkgo biloba extract on the aryl hydrocarbon receptor(Ah R) expression.Methods: 32 healthy male SD rats were selected and randomly divided into four groups. They were normal group, model group, EGb low dose group and EGb high dosage group; The normal group rats were anaesthetized open directly from liver tissue and venous blood, model group after anesthesia open separation of superior mesenteric artery, with hemostatic clamp closed mesenteric artery root 30 minutes, then open 60 minutes to form the model of ischemia reperfusion, EGb administration group of penile dorsal vein injection Shuxuening injection 1 hour after the production of ischemia reperfusion model. The samples were collected and examined as follows: 1 The alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected in each serum. 2 Under light microscope morphological and pathological changes of liver tissue. 3 Biochemical methods to examine the changes of the aryl hydrocarbon receptor(Ah R) and malondialdehyde(MDA) in liver tissues. 4 Western-blot method to detect the liver tissue Ah R level. 5 The expression of AhR was detected by real-time fluorescence quantitative PCR.Results:(1) Compared with the normal group, ALT and AST in serum of model group increased significantly; compared with the model group, ALT and AST in high dose group decreased significantly, with statistical significance.(2) The liver pathological changes: compared with the normal group, the liver tissue of the model group are not regular, the central vein and hepatic sinusoid congestion is serious, local necrosis; With model group, low dose group somewhat alleviate congestion, no other significant changes in the case of high-dose treatment group significantly reduced congestion, reduce inflammatory cell infiltration.(3) Detection of MDA biochemical method: compared with the normal group, model group MDA was significantly higher, compared with the model group, high dose group MDA decreased significantly, with statistical significance.(4) Western blot detection method of liver tissue in Ah R rats: compared with the normal group, the content of Ah R in the model group increased significantly; compared with the model group, high dose group can significantly reduce the content of Ah R, and there is statistical significance.(5) The expression of real-time fluorescence quantitative Ah R: compared with the normal group,the model group Ah R content was significantly increased, compared with the model group, the large dose group Ah R significantly reduced, with statistical significance.Conclusion: Intestinal ischemia and reperfusion can lead to the injury of small intestine and liver, the pretreatment of Ginkgo biloba extract can significantly reduce the content of Ah R, which has protective effect on intestinal ischemia-reperfusion injury in rats.
