Part 1 Impact of Helicobacter pylori infection on the expression of human serum and gastric Nucleobindin 2/Nesfatin-1AIM: Some studies have shown that infection can affect the expression of gastrointestinal neuropeptides, NUCB2/nesfatin-1 is a kind of gastrointestinal neuropeptide,which can inhibit appetite and have other functions, there is no study investigated the influence of Helicobacter pylori infection on NUCB2 / nesfatin-1 expression.To investigate the impact of Helicobacter pylori(H.pylori) infection on the expression of serum and gastric Nucleobindin2(NUCB2)/Nesfatin-1 in humans.METHODS: Serum and mucosae were collected from Eighty-three asymptomatic volunteers who received gastroscopy and H.pylori infection was confirmed by 13 C breath test, rapid urease test and Warthin-Starry staining. The objects were divided into H.pylori positive group and H.pylori negative group. The serum NUCB2/nesfatin-1 concentration was measured by enzyme-linked immunosorbent assay(ELISA) and the expression of NUCB2 m RNA in gastric mucosae was detected by Real-time quantitative polymerase chain reaction.RESULTS: The serum NUCB2/nesfatin-1 concentration in H.pylori positive and negative groups did not differ significantly(2.298 ng/mL ± 0.275 ng/mL vs 2.267 ng/mL ± 0.201 ng/mL, P > 0.05). But the expression level of NUCB2 m RNA in H.pylori positive group was significantly higher than that in H.pylori negative group(1.336 ± 0.324 vs 0.914 ± 0.171, P < 0.01).CONCLUSION: H.pylori infection induced the upregulation of NUCB2 m RNA in gastric mucosa, but had no effect on serum NUCB2/nesfatin-1 concentration.Part 2 Impact of Helicobacter pylori infection on the expression of mice serum and gastric Nucleobindin 2/Nesfatin-1AIM: To investigate the impact of H.pylori infection on the expression of serum and gastric NUCB2/Nesfatin-1 expression in mice. Compare differences NUCB2/nesfatin-1 expression in different duration of infection in serum and gastric tissue of H.pylori infected mice.METHODS: C57BL/6 mice were randomly divided into 3 groups infected and 3 control groups,H.pylor infection mouse model were established by using live bacteria suspension oral gavage, control mice were oral gavage with a sterile fluid,2 weeks, 8 weeks and 16 weeks after infection, one infection group and control group mice were sacrificed respectively. The serum NUCB2/nesfatin-1 concentration was measured by ELISA and the expression of NUCB2 m RNA in gastric mucosae was detected by Real-time quantitative polymerase chain reaction.RESULTS: After two weeks, 8 weeks and 16 weeks of infection, serum NUCB2/nesfatin-1 concentrations in infected and control groups were not significantly different. RT-PCR suggested that after two weeks, 8 weeks infection, mice gastric tissue NUCB2/nesfatin-1 expression did not change significantly compare with the control groups,but after infected 16 weeks infection, mice gastric tissue NUCB2/nesfatin-1 expression was significantly higher than the control group. Immunohistochemistry and immunohistochemistry fluorescent showed, H.pylori infection in 2 weeks and 8 weeks, the NUCB2/nesfatin-1 IR of gastric mucosa was no significant difference campare with the control group.but the infected mice gastric NUCB2/nesfatin-1 IR was significantly more than the control group after 16 weeks infection.CONCLUSION: H.pylori infection had no significant effect on the concentration of serum NUCB2/nesfatin-1, short-term infection(2 and 8 weeks) had no significant effect on the expression of mouse gastric tissue NUCB2/nesfatin-1, but the long-term infection(16 weeks) can result in significant increases of gastric mucosa NUCB2/nesfatin-1 expression.
