Objective: To explore the effects of isoflurane on the self-renewal capacity and differentiation of human embryonic stem cells.Methods: Human ESCs was equally randomized to two growp at the first day after cell-passage. Isoflurane group was exposed in 2% isoflurane, 5% CO2, 21% O2 for 6h, while the control group was exposed to 5% CO2, 21%O2 for 6h. Observe the growth of cells, and detect the m RNA levels of stemness markers and marker-genes of three germ layers of ESCs in two groups.Results: At the fifth day, the cell colonies show difference between two groups. Compared with control group, the number of human ESCs in isoflurane group was distinctly reduced(t=3.40,P<0.01). The m RNA levels of stemness markers(Nanog,Sox2,Oct4,Lin28) of isoflurane group were significantly lower than them of control group(t=2.82~3.89,P<0.05). Moreover, the m RNA levels of marker-genes of three germ layers(AFP,T,Nestin) of isoflurane group were showed significantly increased compared with control group(t=2.67~3.33,P<0.05).Conclusion: Isoflurane inhibits the self-renewal capacity of in-vitro human embryonic stem cells and urges them to differentiate in advance. |
