Coagulation factor Ⅶ applies not only to the congenital and acquired the FVII deficient patients, but also can be used in the treatment of FⅦ and FⅨ inhibit antibody of patients with hemophilia. Coagulation factor Ⅶ can be used in bleeding and bleeding prevention of patients with thrombocytopenia and platelet dysfunction platelet disorder,etc. Coagulation factor Ⅶ also can as widely hemostatic agent for ordinary patients of traumatic hemorrhage of hemostasisl. But the traditional method from plasma extraction yield of coagulation factor Ⅶ is very limited, not only production cannot meet the market demand, but also the existence of certain security hidden danger.Firstly, we obtain high expression of coagulation factor Ⅶ (rhFⅦa) stable recombinant CHO cell line protein, the serum free suspension culture was realized on the basis of the suspension and domestication of recombinant cell strains. Through the optimization of culture medium and cultivation technology research, Establishing efficient rhFⅦ with no serum reactor suspended fed batch culture process, protein expression amount reached lmg/L. Then,we preliminarily studied the purification process of rhFⅦa and established the purification process of rhFVIIa by anion exchange chromatography.
