| Primulina ebumea is belong to Gesneriaceae family. Primulina eburnea is also called shisanqi and shihu. Primulina eburnea was born in an altitude of 100-1500 m liestone mountain forest or river bank. It distributes mainly Guangxi, Guangdong, Hunan, Hubei, Sichuan and Guizhou Province. In recent years, medicinal value; effective identification and analysis;in vitro culture; rapid propagation and physiological stress and so on all, but the research at the molecular level to the Primulina eburnea is less. In this paper, from the molecular level, to the Primulina eburnea for high-throughput genome-wide DNA sequencing, developing microsatellite marker, primer design, so that a more comprehensive discussion of Primulina eburnea the genetic diversity.This experient using high throughput genome sequencing technology successfully developed SSR primers, and choose better polymorphism of 12 pairs of primers (LY34, LY66, LY72, LY77, LY83, LY89, LY133, LY151, LY158, LY181, LY245, LY248) for 15 population 430 individuals of genetic diversity research.12 pairs of primers for the detection of the 200 alleles, the scope of the number of alleles (Na) to 13~21, with a mean of 16.7. Effective number of alleles (Ne), the range of 3.107~7.581, with a mean of 6.07; Observed heterozygosity(Ho)range of 0.494~0.839, with a mean of 0.616; Expected heterozygosity (He) change in the range of 0.678~0.868, with a mean of 0.822.Population level, each population average number of alleles (Na) is 5.99, the effective number of alleles (Ne) 3.51, observed heterozygosity (Ho) and expected heterozygosity (He) are 0.617 and 0.675, respectively. Guangxi GongCheng Hongyancun (HYC) one of the highest population genetic diversity (Na= 6.583, Ne = 6.583, Ho= 0.621, He= 0.721), Guangxi YangShuo(XL) population had the lowest levels of genetic diversity (Na= 5.333, Ne= 5.333, Ho= 0.593, He= 0.658). Genetic differentiation (Fst) 0.129, namely there are 12.9% of the genetic variation between populations, up to 87.1% of genetic variation within populations.AMOVA level, Primulina eburnea population genetic variation mainly exist in the population, accounting for 76% of the genetic variation, only 24% of the genetic variation between populations, shows that the genetic differentiation between species is very small.Bottleneck effect of the test results show that, Guangxi Yanshan shoudan (SD) populations and Guangxi Liuzhou Gutingshan (GTS) populations in the P value was 0.02124 and 0.02124, respectively (P< 0.05 or P< 0.01), the bottleneck effect, but the other 13 population P values were greater than 0.05, show that no mutation -- the divergence drift balance, namely no recent experience bottlenecks.Mantal test showed that the Primulina eburneais closely relative to the genetic distance and geographic distance, r= 0.0625, which is that elevation geographical factors affecting cattle Primulina eburnea diversity, genetic diversity are positively related with altitude, negative correlation with the longitude and latitude and longitude.Primulina eburnea showed higher genetic diversity and low level of genetic differentiation, genetic variation mainly occurs within the population. Gene exchange between species is frequently, may be due to the geographical isolation factors, lead to two species through the bottleneck. |
