Objective: The effect of nuclear radiation on blood biochemical parameters, SOD and GSH-PX activities in liver, the amplification fragment length polymorphism(AFLP) marker were measured to further clarify biochemical toxicology effects of the nuclear radiation on laboratory red crucian carp, to discuss the feasibility of laboratory red crucian carp used for research on ecotoxicology, and to establish the biomarkers monitoring the radiation pollution.Methods: Laboratory red crucian carp was used as the experiment animal to determine median lethal dose of nuclear radiation. The animal were randomly divided into five groups, four different doses(1/16 LD50、1/8 LD50、1/4 LD50、1/2 LD50) of nuclear radiation treatment group and a control group. After the radiation, serum, liver tissue and blood DNA of laboratory red crucian carp were collected to analysis the blood biochemical, the activity of SOD and GSH-PX, the molecular markers, the gene polymorphism information content(PIC) and Nei’s gene diversity(H) and Shannon’s Information index(I), respectively. After experiments, SPSS18.0, Quantity one, Pop Gene, Origin 8.0 software was applied to analyse data.Results:1. In the acute toxicity test, The LD50 of nuclear radiation on laboratory red crucian carp in 30 d is 31.05 Gy.2. Detected the biochemical indexes of laboratory red crucian carp after expousured in different doses of nuclear radiation, we found that the determining serum enzymes showed a clear dose-effect relationship and time-effect relationship.3. With the doses of nuclear radiation increased,the activity of SOD and GSH-PX was decreased.As time goes on, the activity of SOD and GSH-PX was increased but is still below the normal value.4. The gene polymorphism information content(PIC), Nei’s gene diversity(H) and Shannon’s Information index(I) in normal laboratory red crucian carp is 45.20%, 0.19, 0.28. After nuclear radiation treatment, they are 72.27%, 0.28 and 0.41 respectively, which showed the genetics index rise.Conclusion:1. The nuclear radiation has a toxic effect on laboratory red crucian carp. The LD50 of nuclear radiation on laboratory red crucian carp in 30 d is 31.05 Gy. The 95% confidence interal is 25.94 to 37.15 Gy.2. After expousured in different doses of nuclear radiation, we found that the serum enzymes, SOD and GSH-PX activities in liver showed a clear dose-effect relationship and time-effect relationship. And in a certain range, radiation dose and duration are positively related to toxic effects.3. After nuclear radiation treatment, the gene polymorphism information content(PIC) and Nei’s gene diversity(H) in laboratory red crucian carp are higher.4. The laboratory red crucian carp can be used for research on ecotoxicology.
