ObjectiveIn twenty-first Century,, more and more scientists begin to research genomics with the human genome project, gene scientists used a variety of methods to identify gene complex characteristics, the common features of these methods is analyze each gene individually, repeatedly to collect genetic material, analysis of gene. So far, geneticists have identified more than 1000 diseases associated with SNPs. However, how the SNPs i to influence the disease is not still clear, so its clinical significance is not significant, and the whole process is time-consuming, high cost, in order to avoid the expensive like "milk", an alternative methods have been proposed:This is the establishment of a reference gene group and gene mine, mine is through a combination of gene hybridization between 8 maternal genes of different system itself, have 1000 kinds of highly conserved inbreeding lines. The advantages of mine is to bring the gene diversity and gene mapping function maximization. This topic with mineral rich genetic diversity and the strong mapping function, we can quickly find the target gene, disease model better, the development of new treatment methods of the disease.My topic is the advantages of using the gene to study on frontotemporal dementia (FTD), as well as Inflammatory Bowel Disease(IBD)Method1 The clipping respectively containing 8 maternal chain mouse tail, phenol and chloroform extraction DNA through rat tail tissue, and then these DNA of 8 rat tailmouse through the gene chip sequencing. All sequences of the 8 maternal chain mouse, through a combination of eclectic method through cross breeding more than 20 generation to construct the gene mining.2 in vivo in cytoplasm of normal C-tau protein as detection object, taking Gapdh as the reference, the WT mice, FTD mice and FTD were compared with 3 protection chain after hybridization of these 5 groups of C-Tau protein, to observe whether there are differences.3 The control group and constructed by gene mine in IBD of the intestinal epithelial cells were stained with alcian blue PAS. To observe the number of goblet cells and infiltration of lymphocytes in both cases, the number of goblet cell between whether there are differences in statistics.Result1 the 8 maternal chain in random mating, the formation of 1680 independent F2 hybrid group (G3), the statistics of these 8 maternal gene percentages of gene content in G3 generation, the statistical results showed that:N=3, p>0.05,It is no significant difference between the 3 groups,the G3 contains 8 maternal genetic resources equal distribution chain.2 Controls were FTD and FTD mice and 3 protect the chain after hybridization of these 5groups of C-Tau protein, after statistics showed that there are obvious differences between the 5 groups of C-tau,FTD and protein content of FTD and the 3 protection chain after hybridization (n=3,p<0.05).3 observation and statistics of Inflammatory Bowel Disease (IBD) the number of goblet cells and intestinal epithelial cells, after examination, n=3,p<0.05,found that the number of the number of IBD in the model of goblet cells was significantly less than that of normal epithelial cells.ConclusionThe advantages of mine is to bring the gene diversity and gene mapping function maximization. By means of gene ore rich diversity and powerful mapping function, we can quickly find the target gene, disease model better, the development of new treatment methods of the disease.
