Collecting healthy Poa pratensis in University campus and Xining West Mountain,13 or 10 strains endophytic fungi were isolated by using grinding,Tieback its experimental verification are endophytes. Study on the root, stem, leaf found different parts of its infection rate varies,according to the test and found that parts of the carrier rate:stem>leaf>root. By germination test, growth-promoting screening test strains with strong growth-promoting effect of the number of strains ZH-I isolated from stem of Poa pratensis from University campus..On the basis of a single-factor test, orthogonal test, central composite design of experiments, the research findings of the fermentation, the optimum medium was: starch 25 g·L-1, peptone 6 g·L-1, NaCl 2.5 g·L-1, MgSO4 0.5 g·L-1, initial pH of 6. Optimal conditions were: fermentation temperature is 25 ℃, amount of inoculation 6,rotation speed 160 r / min, t liquid volume 100 mL, fermentation time 6d. According to the optimum medium and cultured environment, repeat the test five times, the biomass obtained was 9.95 ± 0.01 g·L-1.In the fermentation kinetics of the fermentation process based on dry weight of mycelium, changes in pH, dissolved oxygen changes, can determine the strain ZH-I mycelial fermentation endpoint was 6d.After configured mycelium dry weight concentration 7,8,9,10 g·L-1 ZH-I strain broth soaking bluegrass seed, while sterile water soaking as comparison strain ZH-I concentrations of the dry weight 9 g·L-1, bluegrass seed germination rate and shoot to promote the role of the most significant, very significant differences with the comparison.With three different ways of soaking, embedded, instilling roots inoculated into sterile seed inoculation, three inoculation methods are on the plant height, fresh weight, shoot dry weight, root dry weight showed some the growth-promoting effect. Among growth-promoting effect of embedding vaccination is most obvious, and obviously very significant difference from the comparison, followed by soaking, and finally the instilling roots.Co-culture on the plant height, fresh weight and dry weight showed some growth-promoting effect. The fresh weight and plant height are the most obvious, and comparison significant difference is very obvious; Stem dry weight and root dry weight of plants difference was not significant, it may be plant stems and root dry weight very much lower.
