Along with the progress of science and technology and the improvement of people environmental protection consciousness, pectin enzyme with its own advantages in food, pharmaceuticals, paper and textile and other fields play an important role, pectinase demand continues to rise. However, the current domestic pectin enzyme to produce force is insufficient, enzyme activity is not high, the overall quality is low, the market is in short supply situation. So, an efficient highly active pectinase to produce the fungus is particularly important, fermentation condition also needs to be optimized. A pectinase producing strain LYS-5304 was screened from wild ramie soil by methods of plate isolation with Congo red staining. Strain LYS-5304 was identified as Erwinia carotovora according to morphological observation and 16S rRNA sequencing. Furthering studying, by means of single factor experiment and response surface analysis, showed that optimum conditions for pectinase production were as follows:Culture media containing glucose 3.0%, rapeseed meal 0.2%, inoculation volume 1.5%, initial pH of 8.0, at 37℃ shaking at 180 r/m for 4 d,.The activity pectinase of LYS-5304 reached 1021.33U/mL under these conditions, and its indicated some potential applications. Enzymology characteristics of the test results show that the enzyme is the most optimum pH range between 8 and 9, belong to alkaline pectinase. The enzyme of the optimal reaction temperature is 50 ℃, the optimum reaction time is 30 min. Metal ion Cu2+ and Ca2 + activation of enzyme protein. At the same time, the strain in maintaining high pectin degradation ability, on the basis of not cellulose enzyme activity, this for ramie degumming has extremely important significance in the textile industry, because it can not only accomplish traditional degumming, and also can not destroy of the ramie fiber composition, maintaining fiber flexibility, therefore has great research value, can consider to further in-depth study.
