Common reed Phragmites communis Trin. is a graminaceous plant and widespread from lakeside to desert regions. Phragmites has adapted to adverse terrestrial habitats such as saline meadow, rocky places and desert dune places. Recently there have been many reports on development of drought resistance molecular mechanism and drought resistant proteins of plant. Phragmites, one of anti-adversity and pioneer plant, include Swamp reed and Desert dune reed. We separated and indentified the proteins of Desert dune reed that response to drought through proteomics, and bring forward some new genes and proteins that probably response to drought of Graminales(Poales).We measured several physiological indexes of Phragmites. We found the activity of POD, content of Proline were increased when Phragmites under dehydration. They are the evidences that Phragmites is highly tolerant against drought. Through 2-DE, proteome maps were constructed for the Phragmites under standard growth conditions, dehydration . Totally, 59 proteins that were changed more than 2 folds were analyzed by MALDI-TOF-TOF MS/MS. Now 25 known proteins and many unknown proteins were identified with database searching.By comparative proteomics, the conclusion could be drawn that laboratory dune reed can study the single lab stress such as drought, and the mechanism of reed that response to drought is probably through the calvin circle in dark reaction of photosynthesis and protein biosynthesis.The up-regulation of sedoheptulose-1,7-bisphosphatase and thioredoxin peroxidase is essential in desert dune reed. Some proteins that vary in natural conditions do not change in drought tolerance.
