Grifola umbellate is a traditional Chinese medicine mushroom, and it has been popularized for its antitumor activity, immune-modulation and other medical efficacy. But the maturation of sclerotia needs a long time about three year, the problem of demand exceeds supply was raised. This paper study on the solid-state fermentation, which could fast valued-added mycelium to solve this problem. To promoting the industrialization of P. umbellate solid fermentation, this study was focus on the optimization of the submerged fermentation process and conditions and the solid process and medium. The main conclusions were as follows:(1) The optimization of submerged culture medium could effectively improve the biomass of P. umbellate submerged culture. The results were as follows:The optimal conditions of submerged culture of P. umbellate mycelium growth were: culture medium with starch 3.0%、soybean meal 3.5%、KH2PO4 0.1%、MgSO4·7H2O 0.1 % and VB10.01%; pH at 6.0、broth content at 40%(v/v)、cultured at25℃for 7d, biomass would reach at 1.97g per 100ml medium.(2) The optimization of solid culture medium could effectively improve the productions yield of P. umbellate solid polysaccharide.The result was as follows:The optimal conditions of solid culture of P. umbellate mycelia polysaccharide were: medium with cornmeal 60%、wheat bran 30%、starch 10%、Armillariella extract 5%、KH2PO4 0.1% and MgSO4.7H2O 0.1% and Fecl3 0.1%, Inoculation amount at 40%(v/v)、Amount of water at 100%(v/g)、cultured at 25℃for 16d, polysaccharide amount would reach at 1.413mg per 1g medium.(3) To improve the yield of grifola umbellate polysaccharide fermentation, this paper study on impact of Armillary for the grifola fermentation. The main conclusions were as follows:The optimal conditions of solid culture of Armillary mycelium growth were:culture medium with cornmeal 80%、wheat bran 20%、KH2PO4 0.1% and MgSO4.7H2O 0.1%; Inoculation amount at 20%(v/v)、Amount of water at 120%(v/g)、cultured at 27℃for 16d, biomass would reach at 15.76g per 100g medium.When the dosage 5% of Armillary solid mycelium extracts added into the grifola umbellate solid medium, its can significantly improve the yield of grifola umbellate polysaccharide, which could reach at 13.4%.(4) Optimum design of isolation of grifola umbellate solid mycelium polysaccharide (GUSFMP) used orthogonal tests. GUSFMP was extracted in boiling water.Removed starch by methods of enzyme. Removed protein by methods of lead acetate. GUSFMP was precipitated in different ethanol concentration. and then it was separated and purified by SephadexG-100 column chromatography. GUSFMP was proved to be pure through column chromatography on G-100 and UV spectrum. The composition of GUSFMP was analyzed by using IR spectrum. The molecular weight of GUSMP was determined by gel penetration chromatography. The result showed that the optimum conditions of isolation of GUSFMP is 5h,temperature at 90℃, shaking and sample/water being 1/50.30%concentration of ethanol GUSFMP was uniform. The molecular weight of GUSFMP I30 was 23710 Da.The study aimed at the aptimization and the controllability of solid fermentation process of P. umbellate, and improved the technology of solid fermentation, through the research of the conditions and technical parameters of fermentation. It would provide theoretical basis for the fermentation production of P. umbellate or other medicine mushroom.
