The Grifola frondosa is a kind of edible and medicinal fungi and it is very rare.Grifola frondosa is various of nutrition which is more than the other edible or medicinal fungi.Grifola frondosa has many biological activities including anti-cancer,anti-oxidation,anti-tumor,anti-viral,immune regulation,the treatment of hepatitis,lowering effects of blood fat and blood sugar and so on.For this reason,the Grifola frondosa has a high medicinal value.In this topic,we extract the antioxidant ingredients from the Grifola frondosa sporocarp.Then,we explore the antioxidant ingredients preliminarily in means of the HPLC and mass spectrometry.1.We tried to extract the antioxidant ingredient from the Grifola frondosa and we measured the total reducing power as parameter.We find the optimum extraction process by means of single factor and orthogonal experiments.When the ratio of solid and liquid=1:50,extracting at temperature of 70? for 2.5 hours,we get the best extraction result.2.The antioxidant ingredients from the Grifola frondosa are mainly the polysaccharides and the polyphenols.We purified the polysaccharides by means of column chromatography of DEAE Sepharose Fast Flow and we purified the polyphenols using the macroporous resin of NKA-?.After the process of purifying,we get the different samples from the Grifola frondosa.There are four samples from the column chromatography of DEAE Sepharose Fast Flow.They are named as GFP I,GFP?,GFP? and GFP?.There are three sample from the purifying process of macroporous resin of NKA-? and They are elution ingredients of 20%ethanol,40%ethanol and 60%ethanol.3.This paper compared the different antioxidant ingredients from the Grifola frondosa by means of DPPH clearance,hydroxyl clearance and the total reducing power experiment.The result showed that the polyphenols have better antioxidant effectiveness than the polysaccharides from the Grifola frondosa and the purified polysaccharides show little antioxidant effectiveness.The antioxidant experiment of different polyphenols from the Grifola frondosa show that the elution ingredient of 40%ethanol play a important role in antioxidant effectiveness.4.After the experiment of different mobile phase system and the different elution time procedure,we find the best HPLC analyzing condition.Then we analyzed the different polyphenols from the Grifola frondosa by means of HPLC.The result is that the medium polar polyphenols from the Grifola frondosa may play a more important role in antioxidant effectiveness.5.This paper combined the HPLC and mass spectrum to identify the antioxidant polyphenols in the Grifola frondosa.In way of compare the spectrogram of DAD and mass,we can infer some antioxidant polyphenols from the Grifola frondosa.They are marlic acid,Coumaric acid,resveratrol,caffeic acid,4-hydroxy-benzoic acid and so on.
