[Armillaria mellea (Vahl ex Fr.) Quel] is a kind of famous medicinal and edible Fungus Belonging to [Armillaria(Fr.)Staud.], Tricholomataceae. Many functions were found in Armillaria mellea polysaccharide such as radioprotective effects, promoting hematopoietic, inhibition of tumor growth and regulation of immune function. Studies of Armillaria mellea chemical Constituents were centralized on fruiting body, rhizomorph, mycelium, etc. there were a few researches on broth. In order to discover the effective Compone-Nts of Armillaria mellea, illustrate deeply the medicinal value of Armillaria mellea polysaccharide, and take full advantage of Armillaria mellea polysaccharide resources, in the paper, the polysaccharide was separated, extracted and purified from fermentation broth of the Armillaria mellea. Homogeneous polysaccharides were obtained from it and their structures were studied.It is established experimental foundation for further research and utilization of Armillaria mellea fermentation broth.The main contents and conclusions in the research were as follows:(1) Extraction process of polysaccharides from the fermentation broth of Armillaria was optimized. Extraction rate of Armillaria mellea polysaccharide was used as a measure. On the basis of single-factor test, orthogonal test was used to optimize the selections. The best extraction technology was that Armillaria mellea fermentation broth and water were mixed as1:3, stirred-extracted three times, each extraction for6hours.(2) Armillaria mellea polysaccharide was separated with ethanol precipitation method. A suitable alcohol precipitation concentration was determined and crude polysaccharides were separated from the fermentation broth of Armillaria mellea.(3)The aforesaid crude polysaccharides were separate with further fractional separation. Three homogeneous polysaccharides were obtained from the separation of the crude polysacchari-de B adopted sephadex chromatography method, it is respectively AMFP-Ⅰ, AMFP-Ⅱ and AMFP-Ⅲ; The molecular weight molecular, weight distribution and purity checking were tested by HPLC method and the data was processed by application-specific polysaccharide GPC software. The results were that the three kinds of homogeneous polysaccharides were the peak symmetry and sharp peaks and the peak molecular weight of812939D,596217D,27321D. PMP column derivatization method was used to test Monosaccharide composition, Three homogeneous polysaccharides are mainly composed by six kinds of monosaccharide such as the Man, Rha, GalUA, Glc, Gal, Xyl. The structures researches of homogeneous polysaccharides were adopted some related tests such as partial acid hydrolysis, methylation, UV,IR, NMR to explain preliminarily the structure of the three homogeneous polysaccharides.(4) Biological activity of the treatment of vascular neuralgia was studied to Armillaria mellea polysaccharide. Chemical and thermally stimulated method, head-breaking and mice were used to judge the effects of analgesic and protection roles to cerebral ischemia in the component of big molecule of polysaccharide, observing the anti-hypoxic effect of Armillaria mellea polysaccharide part to mice under hypoxic conditions.
