Wangyang ditch belongs to Fuyang River, mainly accepted industrial wastewater ofhigh-tech industrial and economic-technological development zone of Shijiazhuang City,which has been polluted seriously. To investigate the relationship between bacterial diversityand pollution levels in Wangyang ditch and seize the strain degrading pollutants, water andsediment samples of six sampling sites were collected, and the diversity of culturable bacteriawere studied in spring, summer, autumn of2013. Bacteria of all samples were isolated andcultured with TSA medium, and864bacteria were obtained, through DNA extraction, PCRand sequencing, and then the results were compared in NCBI, the strains which homology of16SrDNA was greater than97%were defined as same strain. The results were followed as:1864bacteria were obtained and the16S rDNA sequence of them were measured,phylogenetic analysis was done. The results showed that the samples of water and sedimentsin six sites contained seven groups, the water contains24species, and the sediment contains21species, including Proteobacteria (Alphaproteobacteria, Betaproteobacteria,Gammaproteobacteria, Epsilonproteobacteria), Bacteroidetes, Firmicutes and Actinobacteria.The γ-proteobacteria was absolutely dominant group in water, accounting for85.8%of totalbacterial species, the dominant bacteria of Aeromonas for34.8%. In sediments, Firmicuteswas absolutely dominant group, accounting for80.3%in total bacterial species, and thebacteria of the Bacillus was the dominant species for34.2%. Bacterial diversity in water wassignificantly higher than in sediments, and the dominant groups were different.2The bacterial colonies in six water samples tended to gradually decrease in everyseason of2013, which indicated that water pollution became more serious from upstream todownsteam of Wangyang ditch.The bacterial colonies in sediments tended to be stable, exceptthe second sampling site where was near Haoxue flourmill. Because organic matter wereaccumulated largely in mud, and helped the survival of bacteria, the bacteria in the sedimentof the second sample site amounted higher than other sample sites.3The sequence of bacteria colonies was: summer>spring>autumn. The relevant analysis indicated that the bacteria colonies in water were in significantly positively correlated with Hand E, positively correlated with DO and SD, siginificantly negatively correlated with TP,negatively correlated with TN. The bacteria colonies in sediments were in significantlypositively correlated with H, E and Do. Polluted degree of Wangyang ditch affected thebacterial diversity seriously.
