Heavy metal pollution, which causes ecosystem degradation and influences human health, has become one of the most serious environmental problems today in mining. Mine environment contains abundant microbes, meanwhile, microorganisms involve in a variety of element geochemical cycles. To explore the community structure of Linglong Glod Mine and Baiying Copper Mine, the culture method and 16 S rRNA clone libiay were employed in this study.In Linglong Glod Mine, 28 aerobic and 34 anaerocic isolates were obtained from MR sample. 28 aerobic isolates affiliated to three phyla dominated by Proteobacteria, followed by Actinobacteria, and Firmicutes, containing six genera. 34 anaerobic isolates obtained were belonged to Firmicutes and Proteobacteria, dominated by Firmicutes, containing eight genera. Meanwhile, 26 aerobic isolates and 71 anaerobic isolates were obtained from SS sample. Aerobic isolates belong to γ- Proteobacteria, α- Proteobacteria, β-Proteobacteria, Firmicutes, Actinobacter, containg six genera. Anaerobic bacteria belong to Firmicutes, containing seven genera. Compared to cultivation method, 16 S rRNAclone library was built and 48 OTUs were identified from two samples, belonged to six phyla Proteobacteria, Actinobacteria, Bacteroidetes, Acidobacteria, Firmicutes and Planctomycetes, and represented by 36 genera, exhibited a high level of biodiversity. Many unknown and un-cultivated microorganisms were found exsit in the samples.In Baiyin Copper Mine, 62 and 36 isolates were obtained from 1 sample and 2 sample. The 62 isolates from 1 sample belonged to Actinolmcteria, Proteobacteria and Firmicutes, containing nine genera. In B-2 sample, the 36 isolates also were affiliatedto three phyla: Actinobacteria, Firmicutes, Proteobacteria, containing nine genera. The above results show that there were abundant microorganisms in the environment of mining area. The community structure in Linglong Glod Mine is different from Baiyin Copper Mine, it may because the mineral composition, climate, geographical location and the degree of pollution metals are different in the two regions. The isolates in the two regions all belong to Actinolmcteria, Proteobacteria, Firmicutes, indicating that isolates in the three phyla are generally exist in the mine environments.
