In order to investigate the effect of materials in gene activated matrix to geneactivated. Bioactive glass, which is widely used for fabricating scaffolds, is introducedto our study for discuss its role in gene transfection.Sol-gel method is used for preparing bioactive glass sol, polyvinyl butyral issupplied to adjust the viscosity of the sol. Then the sol is processed byelectrospinning to obtain fibers. By control the technological parameters, for example,the molar ratio of distilled water to tetraethylorthosilicate is4~6, the volume ratio ofdistilled water to ethanol is3~5, the aging time for sol is1~3days, the content ofpolyvinyl butyral is3%~5%, we can obtain bioactive glass fibers with less beads,uniform diameter in0.5~3micrometers. If we add Poly(ethyleneglycol)-block-poly(propylene glycol)-block-poly(ethylene glycol) to the sol, such asthe content of2.5%, then adjust polyvinyl butyral to3%~5%, bioactive glass fiberswith hierarchical nanopore are fabricated.For the purpose of stabilize the target gene and real-time monitor the geneexpression, we rebuild a plasmid which can encode fusions of osteoactivin and greenfluorescent protein by recombinant gene technology. The plasmid can replicateautonomously and express target protein.Then we investigate the capacity of bioactive glass fibers in gene transfection.When the content of bioactive glass fibers is1000μg/mL, the content of plasmid is4~8μg/mL, the culture time of before tranfecting is about30min~10h, and tranfect cellswith fetal bovine serum,293T cells can have a good transfection. This character ofbioactive glass fibers mainly due to their good and fast capacity in Ca2+releasing, andrapid co-precipitate to calcium phosphate. They may undergo the same way as calciumphosphate gene nonviral vector by loading plasmid in the co-precipitating process.At last, we use both bioactive glass fibers and lipofectamineLTX_PLUS/plasmidcomplex to transfect MG63cells, the results show bioactive glass fibers a goodapplication in promoting gene delivery and osteogenic.
