Antibiotics,due to its stable efficacy,is used to the medical enterprise and Livestockbreeding widely. Achieved cure for human and animal diseases rapid and effective,effectively control the spread of the epidemic. But uncontrolled use of antibiotics will alsobrings to the natural environment and human health cannot predict the reaction,threat tohuman survival Seriously. So we need to establish a detection means which could separatesuch antibiotics effectively, to achieve the efficient separation, fast extraction, green andenergy saving effect.Small molecular organic solvent aqueous two-phase system as a new separation system,have arouse scholars concern, because of the technique operation simplify, its mild extractionconditions, can not cause the substance inactivation, and small molecular organics cost low,easy to experiment amplification. Applied in the field of biology, medicine, environment,foodstuff field research widely. Small molecular organic solvent aqueous two-phase systemoften adopt unitary system, we can not found the research about binary aqueous two-phasesystem. In this paper we build a new binary small molecule alcohol aqueous two-phasesystem to extract tetracycline class of antibiotics which based on the theory of traditionalaqueous two-phase extraction technique and take advantage of different kinds of smallmolecule alcohol solvent. Provide a new experimental basis for antibiotic residues analysis,test research.In this paper, based on the traditional theory of aqueous two-phase extraction, we basedon: Small molecule alcohol-Salt binary aqueous two-phase system, Investigated the effect ofinfluence factors to oxytetracycline. Confirm system of material composition andproportion,the kinds and concentrations of salt,PH,temperature based on partition behavior ofoxytetracycline in the system. Under the optimum experimental conditions,build the binaryaqueous two-phase system N-Butanol-Ethanol-NaH2PO4,when change the influence factorssuch as the kinds and concentrations of salt,pH,temperature,standing time. We can foundextraction efficiency of tetracycline hydrochloride is94.6%.The result compare with theresearch result that small molecule alcohol aqueous two-phase system extract tetracyclineclass of antibiotics have higher extraction rate.N-Butanol-Ethanol-(NH4)2SO4binary aqueous two-phase system,Investigated partitionbehavior of doxycycline hyclate in the system when change the influence factors such assystem of the kinds and concentrations of salt,pH,temperature,standing time. the amount of antibiotics.Under the optimum experimental conditions,the binary aqueous two-phasesystem, N-Butanol-Ethanol-(NH4)2SO4, to extraction efficiency of Doxycycline Hyclate is92.25%.Through experimental research conclusions that binary small molecule alcoholaqueous two-phase system have advantage such as low cost, small pollution, retrievable,different kinds of small molecule alcohol can decrease the disadvantages each other to form amore perfect as a whole and the binary aqueous two-phase system phase separation into fast,no emulsifying phenomenon, separation effect is better.
