Tetracycline antibiotics is the main source ofantibiotics in the environment, mainly including the City of Tetracycline, oxytetracycline(OTC), doxycycline (DXC), metacycline (MTC). Because of its low content, establishan efficient and rapid method for separation and detection of tetracyclines residuestetracycline antibiotics to reduce the threat to humans and the environment.Based on small molecule alcohol aqueous two-phase system and small organic moleculesin aqueous two-phase system, a simple and new aqueous two-phase extraction composed ofethanol and acetone-Ammonium sulfate binary was developed for the extraction tetracyclineantibiotics. And using the system for tetracycline hydrochloride and doxycycline. Specificresults are as follows:(1) Based on small molecule alcohol aqueous two-phase system and small organicmolecules in aqueous two-phase system, a simple and new aqueous two-phase extractioncomposed of ethanol and acetone-Ammonium sulfate binary was developed.(2) Alcohol and acetone binary Aqueous two-phase extraction separation tetracyclineantibiotics.The influencing factors on tetracycline hydrochloride allocation behavior were alsostudied, including the types,amounts and proportions of salt, alcohol and Small molecularorganic,the pH and the coexisting substances. The results showed that the extraction rate anddistribution coefficient of doxycycline hydrochloride will be up to90%and57, respectivelywhen ammonium sulfate is41%, the ratio of absolute ethyl alcohol and acetone is2:1, thesolution acidity is in pH4.55scope. Allows the relative error is±7%, the K+, Na+, F-,Cl-, NO3–, Br–have no interference on the extraction effect of75mg/L hydrochloric acidtetracycline, the hydrochloride metacycline of1.5times concentration, chloramphenicolpenicillin and erythromycin of35times concentration, the starch and sucrose, glucose of80times concentration and fructose on400times concentration had no interference on results.(3) Alcohol and acetone binary Aqueous two-phase extraction separation doxycyclinehydrochloride. The influence factors on partition behaviors of doxycycline hydrochloridewere studied, including the concentration of (NH4)2SO4, anhydrous alcohol and acetoneconsumption, pH value, temperature and doxycycline hydrochloride. The results showed that: when using pH value of4.5-5.0, The temperature of25degrees Celsius,and (NH4)2SO4concentration of41%, the extraction rate and distribution coefficient of doxycyclinehydrochloride will be up to93.61%and83.081, respectively. Allows the relative error is±6%,the K+, Na+, F-, Cl-, NO3–, Br–have no interference on the extraction effect of80mg/L hydrochloric acid tetracycline, metacycline of2times concentration, chloramphenicolpenicillin and erythromycin of40times concentration, the starch and sucrose, glucose of90times concentration and fructose on400times concentration had no interference on results. |
