With the deep development of reformation, the living conditions have been improvedgreatly. However, the disease gastric ulcer due to drinking or certain reasons occursincreasingly. The disease features on long process, and easy to repeat. The drugs on gastriculcer at the moment include mainly H+receptor antagonists and proton pump inhibitors andgastric acid inhibitors, etc., but the unnatural medicine shall produce some side effects duringthe therapy.Fucoidan (FUC) is a hydro polysaccharide contained in brown algae specially, currentlymost researches on FUC are from Hizikia fusiformis, Ascophyllum nodosum, Laminariajaponica etc. and the study on kelp FUC is relatively rare. As a big country of kelp productionChina takes the first place for about50%of the production in the world; therefore it’snecessary to do deep research on kelp FUC.It is reported that FUC from the algae Capsosiphon fulvescens and Hizikia fusiformishas superior anti-gastric ulcer activity. The kelp FUC belongs to the same components as thetwo mentioned substances, so kelp FUC would probably possess rich physiological functions.The article studies mainly in following two aspects:I. Raise rat by pouring stomach with FUC with different molecular weight, low (300Da-2.5KDa), medium (2.5KDa-10KDa) and high (more than10KDa), in different doses,low (20mg/kg bw) and high (100mg/kg bw). Alcohol generates gastric ulcer model.Through the anatomical lens analysis on gastric sections, stomach tissue HE staining slicing,determination of total acidity and free acidity of stomach, changes on gastric tissues T-SODenzyme activity, MDA, gsh-px and PGE2, to explore the prevention of the components indifferent molecular weight on acute gastric ulcer in the case of different doses.II. Refer to the method of P.C.P. Vasconcelos, to adopt the medium molecular weightgroup, which are stable and strong in preventive ability in acute gastric ulcer model, to studyon the adjuvant treatment in peptic gastric ulcer (chronic gastric ulcer) with three differentdoses, low (40mg/kg bw), medium (100mg/kg bw) and high (200mg/kg bw). In addition tothe traditional control group, there is an experiment of positive drug control group(50mg/kg bw). By gastric anatomical lens observation, stomach tissue HE staining slicing,determination of pH in gastric juice and T-SOD enzyme activity of stomach tissue, MDA, MPO, and NO changes, to explore the adjuvant therapy of medium molecular weightcomponents on chronic gastric ulcer under the condition of different doses. Main conclusionsare as follows:1. Kelp FUC has relatively good preventive effects on irritable gastric ulcer with alcoholcatalytic. Especially the components of both low and molecular weight from the degradationof free radicals have better antiulcer effect than high molecular weight components; themechanism may be that FUC improves the activity of antioxidant enzymes (T-SOD andGSH-Px), inhibits alcohol from lipid oxidation, as well as can increase the content of theregulating factor PEG2in acute prevention.2. The kelp FUC of medium molecular weight7.75kDa has a relatively good assistantcure on the chronic peptic gastric ulcer with acetic acid as the catalyst, mainly by improvingthe activity of antioxidant enzymes T–SOD, inhibiting lipid peroxidation, and gastricsecretion of NO. The activity of MPO in each dose group decreases in comparison with modelgroup insignificantly, and its effect on inhibiting inflammatory cells infiltration is notsignificant.
