Stromata of Shiraia bambusicola, whose effective composition hypocrellin, as the newmedicine of light therapy, has great potential in clinically curing several skin diseases,rheumotoid arthrosis etc such as cancer and cardiovascular disease, is a medicinal resourcesworthy to be developed.The strain that the lab selects to produce hypocrellins pigment has increased greatly inproducing the pigment than other strains reported before. The research based on previousresearch chooses this strain to optimize condition in which the hypocrellins is produced undersolid-state fermentation so as to shorten the fermentation period and increase the pigmentyield. In addition, this research explores its industrialization which lay foundation for theproduction of hypocrellins.First of all, the research optimizes the conditions in which Shiraia bambusicola producehypocrellins under solid-state fermentation and makes sure that the perfect culture conditionis under30℃of fermentation temperatures,55%of water content,2mL·(30g)-1ofinoculation,20g of culture load,(250mL of conicalflask), the original natural pH, onceturnover every other day, and15days of fermentation period.After choosing the perfect culture condition, this research optimizes the componentmaterial of medium by single-factor experiment and orthogonal design. The result shows themedium’s optimal ratio:(0.78-0.95mm)66.8%of corn,16.72%of bran,3.0%of powderedyeast,6.68%of sugarcane straw,5%of glucose,1.5%of sodium nitrate,0.2%of calciumchloride(proportion by net weight). Under this condition, the production of pigment ofconicalflask can reach as high as2.30%.Based on the above research results, this research explores the pilot-scale craftwork ofsolid-state fermentation, employs tray fermentation and studies the controlling methods inbalancing the temperature of material and water content. This research investigates the changeof the temperature of material and water content under the natural situation and controllingsituation. The results show that the material is about8cm thick and we can make use offermentation controlling system to maintain the temperature of material is30℃, the watercontent of material is55%if we control the fermentation conditions. In2days afterinoculation, turnover the culture, and the fermentation period is7-8days, the pigmentproduction up to more than2.74%.
