Compared with traditional steam distillation, steam direction is opposite to pass through the materiel in hydro-diffusion distillation progress, steam pressure and gravity can enhance the extraction effect. Cinnamon leaf as the major raw materials and cinnamon oil extracted by hydro-diffusion distillation method was optimized in a5L stainless still.At the same time, the bioactivity of polysaccharide and flavone as by-products were studied, the results showed that:(1) The extraction conditions of cinnamon oil were optimized by single factor experiment. The optimal conditions of cinnamon oil extracted from cinnamon leaves by hydro-diffusion were as follows:the steeping time is24h, the extraction time is90min, inventory of the material is200g·L-1and the best inlet steam pressure is1750Pa, under these conditions, the extraction rate can reach to8.88‰.While the yield of flavonoids and polysaccharides from cinnamomum cassia leaves were0.039%and0.818%respectively.(2) Compared hydro-diffusion distillation and steam distillation from following aspects:oil yield, extraction speed, steam consumption, drop of steam pressure, Percentage of cinnamal dehyde:①To hydro-diffusion distillation the best material inventory is1000g when the steam pressure is1750Pa, the oil yield is7.498%o;while to steam distillation the best material inventory is850g,the oil yield is7.228%o.The production capacity of hydro-diffusion distillation is22.04%higher than steam distillation,owning faster extraction speed, the inlet steam pressure decreased to51.76%, the percentage of cinnamal dehyde is1.14%higher than the oil extracted by steam distillation.②The essential oil yield can reach to8.88‰when the inventory is1000g and the steam pressure is1750Pa. Under these conditions, hydro-diffusion distillation method is more energy efficient, owning faster extraction speed, the inlet steam pressure decreased to59.57%, the percentage of cinnamal dehyde is2.19%higher than the oil extracted by steam distillation.③The oil extracted by steam distillation is made of four main components: Cinnamaldehyde(84.22%), Benzoicaldehyde(1.1%), Coumarin(3.82%), Metho-xycinamal dehyde(7.07%), while the oil extracted by hydro-diffusion is made of another four components:Cinnamal dehyde (87.89%), Benzoic aldehyde (1.55%), Cinnamyl acetate (3.92%), Methoxy cinamal dehyde(4.56%).(3) The antioxidant activity and antimicrobial activity of cinnamomum cassia leaves flavonoids and polysaccharides were researched. The result showed that the IC50for scavenging·OH rate of cinnamomum cassia leaves flavonoids and polysaccharides, Vc were0.034mg·mL-1,1.605mg·mL-1and0.046mg·mL-1respectively. The IC50for scavenging superoxide’s anion rate of cinnamomum cassia leaves flavonoids and Vc were0.058mg·mL-1,0.028mg·mL-1respectively. The antimicrobial activity of flavonoids and polysaccharides were studied. The results showed that the two objects had apparent inhibitory effect to Staphyloccocus aureus, Bacillus subtilis and E.Coli, the MIC of flavonoids were0.2602mg·mL-1、0.2818mg·mL-1、0.3027mg·mL-1; and the MIC of polysaccharides were3.203mg·mL-1、3.028mg·mL-1、3.375mg·mL-1.
