Lactobacillus plantarum P-8is a probiotic strain with good properties including strong tolerance to acid and bilegood adhesion to gastrointestinal tract, and the beneficial properties of easing blood pressure and blood fat, as well as improving intestinal function are all conformed to the standard of industrialized production. This project was aimed at researching for the laboratory evolution characteristics of L. plantarum P-8by comparing the morphological characteristics, probiotic characteristics and genomic differences between Oth and1000th generations strain.As a result, comparing to the0th generations strain, there is no difference being odserved for the bacterial cell and colony morphology for the1000th generations strain, as well as the favorable bacterial activity decreased even recession phenomenon. In terms of the probiotic characteristics, the tolerance to simulated gastric fluid or simulated intestinal fluid and autoaggregating rate were not changed, while the bile salt tolerance rised slightly. Then the whole genome re-sequencing results of1000th generations strain showed that24mutations were found, and it contained14SNP,3insertions and7deletions.18mutations located in the gene coding region probably caused gene function changed, while the mechanism needed to further research. At the same time, we expected to establish a scientific method to searching for probiotics laboratory evolution characteristics.
