In this study, the strain of Lactobacillus plantarum P-8with0and1000generations were actived after collection in carbon source-limited MRS. Physiological characteristics and probiotic characteristics were decteted, and we analysised the whole-genome re-sequencing for1000genergations.The results showed that after subculturing for1000generations, there are no changes for colony appearance and bacterial cell of Lactobacillus plantarum P-8were observed; the metabolic ability of the bacterial activity of Lactobacillus plantarum P-8was no change after subculturing for1000generations. During Long-term cultivation for1000generations, viable counts and total biomass (OD600) increases with the culture showed changes in volatility, but the total number is unchanged; the tolerance of Lactobacillus plantarum P-8for pH2.5simulated gastric fluid, pH8.0simulated intestinal fluid and bile salt remains unchanged. The auto-aggregating rate was not affected by1000generations subculturing. The analysis of whole-genome re-sequencing for1000generations was showed there were6single-nucleotide polymorphisms (SNPs),6insertions and9deletions. Though SNPs were less, they were high-frequence.Lactobacillus plantarum P-8was cultuered in carbon source-limited MRS for1000generations. The mutations were less, but the physiological characteristics and probiotic characteristics were stable, and no recession.
