Little is known about the nature of buckwheat as medicinal and edible crops, not only have lower blood sugar levels, against oxidative aging effect, but also has the ability to treat cardiovascular and cerebrovascular diseases. However, due to grain contains a large number of highly active anti-nutritional factors and resistant starch and allergens, making the nutritional value of buckwheat greatly reduced. However, germination of buckwheat have improved the nutritional quality of the role. By studying the different absorption time, temperature, humidity dynamic culture change and buckwheat sprouts of each component to determine the best conditions for germination. Using Response Surface Optimization of ultrasonic extraction of flavonoids buckwheat sprouts, and using AB-8macroporous resin was then purified to provide a theoretical basis for the further development and utilization of buckwheat sprouts.The results were as follows:(1)Through the different absorption time, temperature and humidity culture test germination potential and germination rate of buckwheat kernels, buckwheat kernels come wrapped in wet gauze absorbent after12h, at24℃, relative humidity of65%under cultivation highest germination rate reached94.67%.By measuring the number of days for the different components of buds sprouted buckwheat drawn:the germination day8, buckwheat sprouts flavonoids and soluble sugar content reaches the maximum value. Buckwheat flavonoid content of4.92percent, is1.73times the grain; soluble sugar content of25.99%, buckwheat grain is9.77times. VC content on the sixth day to reach the maximum, then began a slow decline. Buckwheat bud germination protein content increased with the number of days showing a downward trend, but the decline marginally.So choose absorption time12h, culture temperature24℃, humidity65%training, training8d buckwheat bud analysis and study next.(2) By single factor test and Design-Expert response surface experimental design, build ultrasonic extraction of total flavonoids of buckwheat germination model to get the optimal use of ultrasonic extraction germination buckwheat flavonoids of81%ethanol concentration, extraction time31min, extraction temperature of75℃, ultrasonic power300W, extraction under this condition extraction rate can reach91.64%. (3) By screening for four macroporous resin AB-8to determine the best buckwheat sprouts purified flavonoids resin models. Use AB-8macroporous resin and purified flavonoids buckwheat sprouts tests to determine the optimum conditions for its purification, ie the sample solution pH of5.0, the sample concentration5mg/mL, sample flow rate2BV/L, with80%ethanol elution at2BV/L, flow rate, total flavonoids purity of62.83%.
