Gelatin is a biopolymer, it is widely used in the medical and medicine industries. Modification is an effective method to improve the functionalities of gelatin. In this work, modification of gelatin by degradation and succinylation was studied systematically. Application of their resulted products was also researched, and according to the experimental results the preparation of succinylated gelatin injection used for plasma substitute were worked out, which will provide valuable theoretical basis and data for its industrialization.Thermal degradation of gelatin was studied systematically. The influences of gelatin concentration, reaction time and temperature on the molecular weights and their distributions for the final products were investigated, and their action rules were disclosed. The study results indicated that thermal catalysis is an effective method for gelatin degradation without impurity introduced in the system.Succinic anhydride was used to modify the gelatin degradation product, and the influences of the amount of succinic anhydride on the reaction were investigated. The structure and properties of the product were characterized by IR, XRD and amino acid analysis. The results showed that the acylation of gelatin degradation product mainly occurs in (?)-NH2of lysine the same as macromolecular gelatin.Applications of the modified gelatin were researched in this paper.According to the rules of thermal degradation and Succinylation modification, and take Gelofusine as the reference, the preparation technique of the plasma substitute was worked out and the appropriate technical parameters were found. Based on IR, XRD and Aa analysis, it was proved that the lab-made plasma substitute had the same composition and crystal structure as Gelofusine. The technical quality of the lab-made plasma substitute, including appearance, pH value, contents of metal ions, content of chlorine, viscosity, molecular weight (Mn), density, conductivity, acylation degree, iso-ionic point, and impurity was tested. It was confirmed that, specification for the plasma substitute obtained by us in the lab reached the requests of the National Drugs Standard "Succinylated Gelatin Injection", which was the same as Gelofusine.The preparation of the plasma substitute with antibacterial activity was studied with exploration. A novel complex of gelatin acylation modification products and rare earth cerium was prepared, and their antibacterial activity was observed elementarily. It was found that the complex possessed of the precious characteristic of antibacterial activity against staphylococcus aureus, which will found a new way for the modified gelatin to use in the medical and medicine industries.
