Picea wilsonii is a kind of deciduous tree in the family of Pinaceae, evergreen woodyplants. It is an excellent plant with Timber, medicinal, ecological and ornamental functions,and it is an endemic plants Chinese. In this article, we analyse the content of polyphenols inthe leaves of Picea wilsonii. We use response surface analysis methodology (RSM) tooptimize the extraction process of polyphenols in the leaves of Picea wilsonii, analyze theinfluences of different tissues and treatments on the polyphenols contents. then we use DPPHfree radical scavenging activity, ABTS free radical scavenging activity and Prussian bluemethod to research its antioxidant activity. The results are as follows:(1)For reflux extraction, The influence of total polyphenol in needles of Picea wilsoniiwere: temperature> ethanol volume fraction> extraction time> ratio of material to liquid.The best process: ethanol concentration42%, ratio of solid to liquid1:61,extraction time5.1h, extraction temperature63℃, regression model to predict the extraction rate of thetheoretical value of up to22.6387mg/g.The extraction of polyphenols was investigatedwith design response surface method.. The results show that the influence of factors on theextraction rate of polyphenols was the order of ethanol volume fraction> ultrasonic wavepower> ratio of material to liquid> extraction time, and when liquid material1:51(w/v),ultrasonic wave power250W, extraction temperature42℃, extraction time46min were theoptimum extraction method. regression model to predict the extraction rate of the theoreticalvalue of up to27.9586mg/g.By using different extracting methods with same sample, theresult shows that the hot reflux process is better.(2)The IC50of DPPH, ABTS for Picea wilsonii polyphenol were0.0778mg/ml,0.0098mg/ml; equivalent of ascorbic acid0.0334mg/ml,0.0081mg/ml, had a certainreduction capacity for the Prussian blue method to measure the Picea wilsonii polyphenolsolution, but lower than that of ascorbic acid.
