In this paper, effects of l-Methylcyclopropene(l-MCP)on the changes of polyphenols contents in different parts of apples during the cold storage were studied. The Red Fuji apples and Huaniu apples were treated respectively with 200,500,1000 nL L-1 1-MCP at room temperature, and the changes of the polyphenols contents in flesh, peel and core of apples during 0~4℃cold storage were measured by high performance liquid chromatography. The best macroporous resin for separation and purification of the apple polyphenols was screened by static adsorption and desorption tests. Single factor tests were carried out to study the effects of technological factors on the adsorption and desorption prosperities of macroporous resin on apple polyphenols, based on which manipulation parameters of macroporous resin for the adsorption and desorption of apple polyphenols were optimized with response surface method. The model of preventive obesity was taken to evaluate the effect of apple polyphenols on the anti-obesity and preventing of nutritional obesity of mice. The experiments mainly reached following results.1. The polyphenols in Red Fuji apples and Huaniu apples mainly contain such as gallic acid, chlorogenic acid, rutin, phloridzin, but both of them don't contain benzoic acid.2. Compared with the control group, the treatment with 1000nL L-1 1-MCP significantly inhibited the decrease of the polyphenols contents such as gallic acid, chlorogenic acid, rutin, phloridzin etc in apples, while the treatments with both 200nL L-1 and 500 nL L-1 1-MCP were found ineffectively. The quality of the apples preferably maintained.3. As to different varieties, the contents of total polyphenols , chlorogenic acid, rutin, phloridzin from different parts in huaniu apples are higher than in the Red Fuji apples. But the gallic acid content in peel, flesh of the Red Fuji apples is higher than that of huaniu apples.4. To different parts, total polyphenols in peel of apples are higher than in flesh and core. The contents of gallic acid ,chlorogenic acid, rutin and phloridzin in the core of apples are higher than in the peel and flesh.5. The results showed that D280 macroporous adsorption resin was the best macroporous resin in apple polyphenols extraction and purification.6. Response surface experiments were conducted to investigate the effects of multifactors on the adsorption preperities of macroporous resin for apple polyphenols. The results indicated that feeding rate, concentration of feed and pH were three key factors and equation between dynamic adsorption amount and the three factors was: Y= 42.672-0.16A+2.38B-0.34B2-1.36C; The optimum adsorption conditions were at the feeding rate of 1.52ml/min, concentration of 2.50mg/ml, pH 4.25. The maximum dynamic adsorption quantity was 44.72mg/g in theory.7. Response surface experiments were conducted to investigate the effects of multifactors on the adsorption preperities of macroporous resin for apple polyphenols. The results indicated that elution rate, elution concentration and eluant volume were key factors and equation between dynamic desorption rate and the three factors was: Y= 75.98-0.75A-12.89B-5.05B2-1.82C2+0.73AB.The optimum desorption conditions was at 0.50ml/min of the elution rate, which elution concentration was 70%, pH=1. The maximum dynamic deadsorption rate was 84.71% in theory.8. Apple polyphenols could prevent the mouse nutritional obesity and anti-obesity effect. The effect of anti-obesity of mouse were influenced by doses of apple polyphenols. The difference was not marked(P > 0.05)when mouse were perfused by low doses apple polyphenols (0.5mg/10gBW).The middle(2mg/10gBW) and high doses(5 mg/10gBW) could influence significantly (P < 0.05)the anti-obseity, lowering total cholesterol, blood lipid and glucose concentration. But the difference were not marked between middle doses and high doses. The difference was not marked (P>0.05) when rats were perfused at morning and at evening, But the effect was better at morning from biochemistry index.
