Synthetic polyamides containing N-methylpyrrole(Py) and N-methylimi-dazole(Im) amino acids which can permeate living cells, bind to DNA sequence with high affinity and specificity and regulate expression of specific genes are a very important kind of chemical copounds. On the other hand, vitamine C which is the must substance of human body is used as intensifier of DNA cleavage. But there are little reports about DNA cleavage of vitamine C. So studies on the synthesis and characteristics of the polyamide to improve its affinity, sequence-specificity and cleaving efficiency are a research project with great academic value.This thesis deal mainly with the synthesis of the polyamide and ascorbic acid conjugates.The content contains:1. The research about the synthesis of the polyamides. A serise of polyamides were synthesized by N-methylpyrrole and N-methy-limidazole. Most of the building blocks have been prepared by a haloform reaction in a simple way without resorted to column chromatography. With the DCC/HOBT, DCC/DMAP coupling reaction, polyamides three have been effectively prepared without amino protection and deprotection. The way of acylchloride has successfully synthesized polyamides four, is very secrit to water.2. The synthesis of the derivatives of vitamine C. Vitamine C is a good DNA cleavage, whose2,3-OH has DNA cleaving activities.We have synhesied a series of derivatives of vitamine C with hydroxy protection and deprotection.3. The synthesis of the conjugates of vitamine C and polyamides. With the BOP/DIEA coupling reaction, we have been successfully synthesized conjugates of vitamine C and polyamides in the ice.4. The test of the new conjugates. We have characterized the new conjugates by1H NMR,13C ESI-MS spectra.
