Broccoli seeds have a very high nutritional value and medicinal value. Glucosinolate and sulforaphane are one of the best natural products with anti-cancer effect.The content of glucosinolate and sulforaphane in broccoli seeds in different regions-even if is the same region-is obviously different. By the in-depth study of broccoli seeds which can be detected GRA but can not be detected sulforaphane, we found that these seeds can not hydrolyze GRA to sulforaphane by themselves.Investigation in separation and preparation of glucosinolate (GRA) in broccoli seeds, confirmed that the water extracted relative to organic solvents such as methanol extraction is superior. With the research of extraction efficiency of different solid-liquid ratio and the judge of experimental results and equipment factors, the best solid-liquid ratio is1:10.Sulforaphane in broccoli seeds were isolated and purified. In the experiment of the solid-liquid ratio influence on the filtration performance, we found that when the solid-liquidratio is1:8, the pH is adjusted to2-3, the hydrolysate’s filter performance is better. In the preparation of crude sulforaphane production we found that when low sample volume (1to3kg) has a low influence on production yield, but when the sample volume is increased, the production yield is significantly lower. The production yield is also become lower when the resin is used more. In the medium-pressure chromatography preparation of high purity sulforaphane we found that compared with preparative chromatography, medium-pressure chromatography can processing more samples. The optimum conditions for the medium-pressure chromatography are also identified. The mobile phase is15%methanol, the flow rate is100mL/min and the sample volume is4g when the purity of sample is high.
