The intaken of cruciferous plant can reduce cancer risk. Glucoraphenin is the predominant glucosinolate in radish sprout, which is an important cruciferous plant in the world. Glucoraphenin can be hydrolyzed by endogenous myrosinase to sulforaphene, which is implicated in anticancer activity. In order to find the best of germination time, biotic elicitor and cooking mathods for radish sprouts, we investigated the effects of germination, biotic elicitor and subsequent cooking methods and cooking time on the levels of glucoraphenin, sulforaphene and anticancer activity of radish sprouts extracts in germination and cooking processes in this research. In addition, we studied united hydrolysis of cruciferous sprouts. ESP protein existed in many brassica sprouts and it can direct the hydrolysis of glucosinolates to nitril, which has no significant anticancer effect. So we hope to improve isothiocynates using the united hydrolysis of cruciferous sprouts in this research. The major results are as follows:1. we found that the levels of glucoraphenin, sulforaphene and the antiproliferation activity of radish sprouts extracts on human lung cancer H1299 and HCC827cells was decreased gradually in line with the germination process. The additon of L-methionine and L-cysteine can induce the formation of sulforaphene in germination process.This research showed that 3-day-old radish sprouts are an excellent source of high content anticancer compounds that could potentially benefit human health.2. The effects of three traditional cooking methods, including boiling, steaming and microwaving on bioactive components and antiproliferative activity of radish sprouts were also studied in this research. We found that for these three methods, even conducting brief cooking time 0.5 min on radish sprouts, the myrosinase activitylost more than 88% and sulforaphene formation decreased by more than 95% and the antiproliferative activity of radish sprouts on human lung cancer H1299 and HCC827 cells was considerably decreased. Thus eating fresh radish sprouts is the best way for obtaining anticancer activity.3. The united hydrolysis of cruciferous sprouts was studied in this research, and found that sulforaphane formation was increased to 2.03,2.32, and 1.95-fold when broccoli sprouts was hydrolyzed with radish, rocket and rape sprouts. Then we detected the level of sulforaphane nitril and found it decreased greatly. We preliminarily proposed the mechanism of the untied hydrolysis of cruciferous sprouts and found that it can promote sulforaphane formation when brrocoli sprouts was hydrolyzed with cruciferous sprouts outside of brassica species or brassica sprouts which had low activity of ESP protein. These results can make people to obtain more bioactive activity from cruciferous sprouts.
