Study On Synthesis And Properties Of Emodin Derivatives

Emodin extracted from rhubarb is a natural anthraquine compound．It hasbroad pharmacological effects and vatiety of biological activities，Such as the roleof diarrhea，anti-tumor effect，anti-mutagenic and mutagenic effect，antibacterialeffect，anti-virus effect．However，due to poor water solubility and high toxicity，It has not yet been used in clinical report．Therefore，in order to get some googanti-tumor activity，low toxicity of emodin derivatives，Many researchers refineand transform emodin．In recent years，Studying emodin derivatives has becomea hot topic in the field of medicinal chemistry．This paper synthesized three series of emodin derivatives，a total of23targetcompounds，with emodin as raw material．These compounds were chatcterized byIR、elemental analysis、NMR、X-ray crystallography and so on．The first serieswith16compounds mainly enhaced the water solubility of Emodinderivatives．These compounds had been tested，by MTT method in vitro activity，and foundthat some compounds showed good anti-tumor activity on Jurkat and K562leukemia cell lines．Their IC50values are about2.8~4.0μmo/L．Most of thecompounds have shown good antitumor activity．In the second series with5compounds， We mainly synthesized some hudroxy-anthraquinone emodinderivatives，researched their infrared spectroscopy and UV-visible absorptionspectroscopy properties，explored the law of these compounds in the spectra，andpreliminary studied the hydroxyl group ptotection and deprotection of emodinderivatives．The third series with2compounds had been synthesized through theprotection of hydroxyl group，NBS reaction，hydrolysis reduction，condensationreduction and esterification from emodin．We found that the oxime ester ofemodin derivatives，with aliphatic hydrocarbons，could not be obtained in theexperiment，but with aromatic hydrocarbons could be．Moreover，A number ofmature synthetic route had been improved in our experiment，We get somesynthetic routes with high yield and short steps，and summarized these inprovedroutes．This paper priliminary research on emodin derivatives，their biological actityand spectroscopy propetties．It is desirable that this paper could bring much better idea into the development of emodin derivatives with goog biological activity andlow toxicity．...
