With the rapid development of aloe industry, more and more aloe vera food appear inthe table, But it may remains aloin and its hydrolysis product which is aloe emodin in thesealoe products, these two substances are both anthraquinone compounds, which have astrong allergic and diarrhea, it will cause damage to human health if there remains excess ofAloin and aloe emodin in the food. Therefore, in order to control strictly of the aloin and aloeemodin residues in food, and protect the health of consumers, it is important to establish arapid determination method of aloin and aloe emodin. The main purpose of this paper is toestablish a rapid detection method of aloin and aloe emodin by surface enhanced ramanspectroscopy and fluorescence spectroscopy, which is a rapid detection method of highsensitivity.First, based on the structure and characterization of aloin molecule, SERS assay wasestablished. By comparing the raman spectrum of experimental and theory calculated, wedetermined the qualitative peak detection of aloin are461cm-1,818cm-1,1229cm-1.Determined the optimum conditions of SERS detection of aloin from three aspects which areenhanced substrate, solvent and pH adjustment, the best condition is Ag colloid as enhancedsubstrate, ulter pure water as solvent, not adjusting pH. The detection limit is5mg/kg ofSERS detection method.Next, choose1,8-two hydroxy anthraquinone as the template because it is similar toemodin in structure, molecularly imprinted polymer was prepared which can be specificadsorption of aloe emodin, and characterizes it by scanning electron microscopy. With thestudy of dynamics, the adsorption isotherm, the deliberately adsoption properties of theimprinted molecule, results show that the best adsorption time of the imprinted polymer is5h,and the specific adsorption effect is very good for aloe emodin.Last, the hydrolysate of aloin which is aloe emodin aloe were detected by fluorescencespectroscopy, the excitation wavelength is490nm, the quantitative peak is the544nm peak,the standard curve equation of aloe emodin concentration and fluorescence intensity isY=15807X-5701.9, R2=0.998, and the detection limit is0.0005mg/kg.Detect the actual samples use the detected method upon, and compare with the result ofhigh performance liquid chromatography, the correlation coefficient is0.9998, so the resultshow that the method established in this study can be applied in repid detection of aloin andits hydrolysis product of aloe emodin.
