In this work, glass chip-based and short capillary-based microfluidic capillary electrophoresis systems with laser-induced fluorescence detection was developed. The ability to separate fluorescein dyes labeled protein was shown.In the first chapter, the developments of the chip-based capillary electrophoresis for protein separation in recent years are summarized. Various chip-based capillary electrophoresis systems for protein separation based on different capillary electrophoresis separation modes are introduced. Furthermore, the approaches to suppress the adsorption of proteins on the surface of microchannel on chips are discussed.In the second chapter, the protein separation on chip-based capillary electrophoresis was performed using a lab-made bioanalyzer. Cy5 labeledα-chymotrypsinogen A and other proteins are separated. Various methods to suppress the adsorption of proteins on the surface of microchannel on glass chips was studied. The LIF detection and the pinched injection mode are employed for sample detection and introduction respectively. Under the modified conditions, sample could be separated in 225s.In the third chapter, a fast capillary electrophoresis system for protein separation was developed based on a short capillary. The LIF detection and slotted-vial array sample introduction system was used. The protein sample was separated using this system which has the advantages of automated operation, fast separation and good repeatability.
