This paper mainly concerns three points:the cultural conditions of caudaside-glycosidase produced by Absidia sp.O90, the control of enzyme reactions and characterization of caudaside-glycosidase.The experiment purificates crude caudaside for getting substrate. After the disposals of ethanolic extraction, lixiviation of macroporous absorbitive resin and purification of silica gel chromatography, the yield of the composition LM in material is0.068%.To get the optimal conditions for producing caudaside-glycosidase, we selecte Absidia sp.O90, and choose15%lixivium of Caudaside as revulsant, culture192hours. And the reactive condition of crude crude caudaside-glycosidase is0.5mg/mL,50℃, pH5.0and the best time of enzyme reaction is36h.And then, caudaside-glycosidase is purified in the course of separation using the material of DEAE-Cellulose DE52cellulose. Furthermore, the purified enzyme activity and characters of the caudaside-glycosidase from microorganism Absidia sp.O90is determined and the SDS-polyacrylamide gel electrophoresis is used. The enzyme molecular weight is about65kDa.The study about enzyme characters of caudaside-glycosidase, the enzyme is steady under the condition of pH3.0-pH6.0, and the optimum condition of pH is pH5.0; the optimum condition of enzyme reaction was50℃, the stability of temperature between20℃~60℃is the best. Caudaside-glycosidase was stable in acidic condition. And it was inhibited by Cu2+, and Na+can activate caudaside-glycosidase slightly when the concentration is high. The Vmax and the Km of caudaside-glycosidase is15.04μmol/L-h mmol/L·h and2.80mmol/L respectively.
