Raman spectra, which utilize the interaction of photon and molecules, can indicate the information of the constitutes and frameworks of molecules, vibrational energy level, rotational energy level characterizations. As every single material has its own ’fingerprint-like’Raman spectrum, this kind of spectra can detect the composition and structure of some material.Raman spectra have been widely used in materials science, environtology, chemistry, biomedicine, archaeology.This paper introduces the theoretical principles and applications of Raman spectroscopy, as well as the research of analyzing the Raman spectra of serum. This paper also introduces hierarchical clustering analysis to classify different kinds of serum. The main contents of this paper are as follows:Firstly, the beginning of this paper summarize the research and development of Raman on detecting breast cancer and other breast disease and serum Raman test. Successively, this paper presents the theoretical knowledge of Raman spectra from two perspective-classical theory and quantum theory, and Raman spectra’s applications.Secondly, this paper introduces two methods of AgNPs preparation-hydrothermal and microwave method. According to the comparison of serum combined with hydrothermal AgNPs and microwave AgNPs, we chose the microwave method.Finally, original data are processed with method of baseline calibration and normalized with725cm-1spectral peak. Then the normalized relative intensity of Raman spectra are analyzed with method of HCA, and the purpose is to explore a new method of screening for breast disease. On the basis of clinical pathologic diagnosis, the result of this screening is that misjudgment rate about both normal people and breast patients is0%. The primary results show that this method of serum-SERS screening for the patients with breast disease is successful, but when it comes to separate different kinds of breast disease, this method still need more research.
