| As people living standard rise, functional foods are becoming main food of the21st century. Thefunctional Xylooligosaceharides have been a kind of important funetional food additive because they havespecial physiological funetion. But now in China the low poly yield and purity of xylose lags far behindother countries. In this paper, three parts of the cottonseed hull study was Xylooligosaccharidesoptimization decoluring, purification and purification of cottonseed hulls and cottonseed hulls XOSpreliminary activity.1. Cottonseed hulls optimization decoluringUsing three different methods of acid precipitation, activated carbon and anion exchange resin tobleach xylan hydrolysis liquid. Through the study on xylan hydrolysis liquid bleaching process,conclusions as followed:(1) The optimum conditions of acid precipitation bleaching was: pH was3.0,temperature was50℃, the processing time in this condition, decolorization rate was49%;(2) compareingthe bleaching effect of the four activated carbon, best type of powdered active carbon was HAC, theoptimum conditions is: activated carbon dosage was2.08g/L, the temperature was47℃, initial pH3.2,under this condition, the bleaching time was51mins, bleaching rate is74.36%;(3) D290resin has a betterdecolorization effect on coarse sugar, its optimum condition of bleaching temperature was45℃, initial pHwas6.5%of the amount of resin added, bleaching time was3h, bleaching rate was78.4%.2. Further purification of XOS by cottonseed hull after bleachingIn this paper, we select Ultrafiltration and Sephadex G-15column for further separation andpurification of liquid XOS, conclusions as followed:(1) the optimum conditions of ultrafiltration operatingpressures was0.8MPa, operating pH was7.0, the operating temperature was23℃, the mass concentrationwas1.0mg/mL, XOS transmittance rate was92.17%at this time;(2) using Sephadex G-15Sephadexcolumn chromatography to seperate XOS crude, get more pure components;(3) The results of UV bandscanning analysis showed that the protein had been basically removed from the LSO dissolvent after beingpurified.3. Preliminary research activity of cottonseed hulls XOSThe antioxidant activities in vitro of XOS from Cottonseed hulls were evaluated by measuring2,2-dipheny-1-1-picrylhydrazyl (DPPH) radical and hydroxyl radical (OH) scavenging activity,superoxide radical scavenging activity (O-2). The results showed that: XOS has a hydroxyl radicalscavenging, free radical superoxide anion and DPPH radical, and the ability to clear the concentration offree radicals and XOS showed a positive correlation. Has the ability to scavenge free radicals VitaminC>Vitamin E>XOS. Finally, physiological functions of xylooligosacchrides were studied by their in vitrocultivation. The results suggested that xylooligosacchrides could unused effectively by S.aureus and E.coli,and proliferation of probiotic lactic acid bacteria. |
