This paper study the extraction and separation technology of polysaccharidefrom Gold lotus, and its antibacterial activity and antioxidant activity, these providetheoretical guidance for the research and utilization of Gold lotus. Conclusions are asfollows:(1) The optimum process conditions of extracting polysaccharide from Goldlotus by hotwater extraction were extraction temperature for80℃, extracting timefor2.5h, ratio of material and liquid for1:25, the extraction yield was4.89%in thiscondition. The optimum process conditions of extracting polysaccharide from Goldlotus by ultrasonic extraction were ultrasonic power for160W, ultrasonic time for80min, ratio of material and liquid for1:15, the extraction yield was5.32%in thiscondition. The optimum process conditions of extracting polysaccharide from Goldlotus by microwave extraction were microwave power for450W, extraction time for2.5min, ratio of material and liquid for1:10, the extraction yield was5.87%in thiscondition.The optimum process conditions of extracting polysaccharide from Goldlotus by ultrasonic-microwave synergistic extraction were ultrasonic power for120W, microwave power for350W, ratio of material and liquid for1:14, extraction timefor3.5min, the extraction yield was6.54%in this condition. From extractingpolysaccharide by hotwater extraction, ultrasonic extraction, microwave extraction,ultrasonic-microwave synergistic extraction, extraction yield by ultrasonic-microwave synergistic extraction was higher than hotwater extraction, ultrasonicextraction, microwave extraction, and ultrasonic-microwave synergistic extractionhave a promising future.(2) The optimum process conditions of removing protein of polysaccharidefrom Gold lotus were trichloroacetic acid concentration for14%, oscillating time for25min, removing protein time for3times. The optimum process conditions ofremoving pigment of polysaccharide from Gold lotus were activated carbon contentfor2.5%, temperature for50℃, adsorption time for40min. (3) Polysaccharide from Gold lotus had strong inhibition effect to bacteria,mould and Saccharomyces cerevisiae, its antibacterial spectrum was wider. The MICof polysaccharide from Gold lotus was6.25%to Staphylococcus aureus andRhizopus, was12.5%to Escherichia coli and Bacillus subtilis, was25%toSalmonella, Aspergillus niger and Saccharomyces cerevisiae.(4) Polysaccharide from Gold lotus had strong scavenging ability toDPPH·、·OH and O2·, and polysaccharide from Gold lotus had good ability ofanti-lipid peroxidation effect.
