Study On Extraction, Purification And Tablets Of Flavonoids From The Stems Of Hops

The stems of hops contains many flavonoids, it has high medicinal value. The stems of hops as hopby-products were abandoned, underutilization. The stems of hops as raw materials in this paper, Theflavonoids changes in the storage, extraction process, purification,production tablets were studied,experiments concluded as follows:(1) Using the three methods of impregnation, ultrasonic, microwave extract flavonoids from the stemsof hops, the optimal conditions of ethanol impregnation method to extract the flavonoids were obtainedthrough response surface optimization: ethanol concentration of41%, solid-liquid ratio is1:14g/mL,extraction temperature of83℃for dipping3h, under this conditionflavonoids content is33.39mg/g. Theoptimal conditions of ultrasonic extraction of flavonoids were: ethanol concentration of65%, solid-liquidratio is1:12g/mL, temperature of54℃extract32min,the flavonoids content is33.97mg/g. The optimalconditions of microwave method to extract the flavonoids were: ethanol concentration of65%, microwavepower for84%, solid-liquid ratio is1:16g/mL, time for34s, under this condition flavonoids content is34.53mg/g. Microwave method was the most suitable to extract total flavonoids from the stems of hopsthrough three methods(2) Using macroporous adsorption resin purify flavonoids crude extrac trom the stems of hops.Comparing with the flavonoids’ adsorption rate and desorption rate from nine kinds of macroporous resinsto choose the suitable one, AB-8macroporous adsorption resin is the most ideal resin, adsorption rate83.33%and desorption rate85.00%. The appropriate conditions were as follow: the sample concentrationwas1.20mg/mL, pH3,70%ethanol as the eluent, diameter height ratio of1:8, the sample quantily was70mL, adsorption flow rate1.00mL/min, the eluent quantily was100mL, desorption flow rate1.00mL/min,after purification of total flavonoids concentration was3.98times of the original.(3) The optimal conditions of preparation of β-cyclodextrin inclusion compounds were obtained throughsingle factor and response surface optimization, took embedding rate as an indicator. The results showed:ratio of core wall1:11g/g, Using11%β-cyclodextrin as wall material,63℃mixing54min,throughverification test, Model prediction and the measured the embedding rate were small. The optimalconditions of production of tablets were obtained through single factor and response surface optimization,took angle of repose, hardness, disintegration time as an indicator. The results showed: the dosage of (4)β-cyclodextrin inclusion compounds33.5%, the dosage of corn starch40%, the dosage of magnesiumstearate3%, the dosage of5%L-HPC7%. Through verification test shows that optimization of thepreparation of tablets with good reproducibility, stability and practical. Under the condition of thepreparation of tablets, the surface is smooth, colour and lustre is uniform.(4) The present study evaluated the quality of the tablets from aspects of character, disintegration time,weight variance, content determination, and investigated the stability of the tablets. The results showed thatindicators accord with tablets quality requirements of2010《China pharmacopoeia》.
