The total flavonoids from hops were determined by spectrophotometer in this paper; the optimal extracting conditions of flavonoids from hops were studied; researches were done about purification and antioxidant of flavonoids from hops.1. The total flavonoids from hops were determined. Four different methods have been used in the extraction of flavonoids from hops soaked in organic solvents was the best extraction method. Solvent, ethanol concentration, temperature, extraction feed ratio, extraction time and the methods of extraction were selected as the single factor test conditions. Ethanol was selected as the extracting solvent after the single factor experiment was made. Ethanol concentration, extraction temperature and the ratio of sample, extraction time and solvent were selected as orthogonal test of four levels of three factors. The results of orthogonal test showed that the best process of total flavonoids extraction was that extraction temperature was80℃, extraction time was60min, ethanol concentration was60%and the ratio of sample and solvent was1:25.2. The purification of flavonoids from hops was studied. It showed that the macroporous adsorption resin polyamide was selected as exclusive separation resin of flavonoids from hops, the adsorption velocity was3mL/min, elution velocity was1.5mL/min, diameter and height rate was1:7.5, the pH was9, the elution solvent was3BV. Then the elution which got from abovt conditions was dehydrogenized by vacuum freezing drying method, the tan powder of flavonoids from hops was collected. Compared with other methods, the content of flavonoids (60.2%) extracted by this method was more stable.3. According to the total deoxidization、the ability of total anti-oxidize and the elimination of DPPH, It showed that antioxidant from hops had a good anti-oxygenation.4. The anti-irradiation activity of flavonoids from hops in mice irradiated by60-Co-y-ray (7.5Gy, at a dose of1.5Gy/min), the mice were randomly divided into six groups, including the control group, the positive control group (400mg/kg, flavonoids from ginkgo) and four experimental groups (0,100,200and400mg/kg, equivalent to hops) for28days. The mice of the five experimental groups were then exposed to the60-Co y-rays and given flavonoids from hops for another3days. The white blood cell numbers were counted. The activities of glutathione peroxidase, malondialdehyde, superoxide dismutase, catalase, spleen index and thymus index were also determined. The results indicated that the flavonoids from hops showed radioprotective activity in vivo, and the mechanism might be the antioxidative activity of the flavonoids and the modulation of the immune system.
