China has the world’s largest cotton planting area, so there are abundant resource incotton seed husk. Hemicellulose content of cotton seed husk was detected about24-28%, withlimited arabinose, which was ideal raw material for production of higher purityxylooligosaccharides. The main research contents of the paper ware as follows: the processconditions for production of xylooligosaccharides were optimized. Besides, the purificationand the prebiotic function of xylooligosaccharide were also studied.Using cotton seed husk as the raw material were studied the effect of different conditionon the production of xylooligosaccharides. The optimal parameters were as follows:processing time in high-temperature was36.0min, the concentration of sulfuric acid0.17%,and enzymatic hydrolysis in pH6.5.The paper also studied the purification of xylooligosaccharides by different kinds ofresin material. The result showed that the optimal were anion resin D301G and cation resin001×7, with98%desalination and96%decoloring.47healthy adults were provided with monosaccharides at the dose of1g/d. And theirfecal microbial level were examined.The soft of IBM SPSS Statistics20was used to analyzesignificant. The results showed: the xylooligosaccharides could promote the proliferation ofBifidobacterium spp and Lactobacillus spp with significantly effects (p <0.01); the sugarscould inhibit the proliferation of the Clostridium perfringens spp, Escherichia Coli spp andEnterococcus spp with significantly effects (p <0.01). In addition, the average of PI value bythe sugars, which estimated the effect of prebiotics were2.15with the minimum value1.32.The results showed that xylooligosaccharides were beneficial to human body.The influences of xylooligosaccharides on fecal short chain fatty acid were also studied.The results showed that the xylooligosaccharides had significantly effects on increasing lacticacid and acetic acid concentration (p <0.05), while there had no significant changes in thecontents of the propionic acid, butyric acid (p>0.05). so the xylooligosaccharides producedfrom cotton seed husk could proliferate human intestinal beneficial bacteria.An efficient bioprocessing method of xylooligosaccharides was studied. The resultsdemonstrated that xylooligosaccharides produced has significant prebiotic effect, selectively increasing Bifidobacterium spp and Lactobacillus spp.
