Background:Rose fruit is the dried ripe fruit of Rosa davurica Pall.,which is widely used inbeverages,health products and other fields currently.Researches have shown that thethe fruit has plenty of chemical components,such as flavonoids, polysaccharides,saponins,vitamin and so on.The pharmacology study indicated that the fruit has thefunctions of promoting digestion,antioxidant,anti-aging,improving the immuneregulation and antiradiation.It has important significant to carry out the foundmentalresearch on the nature fruit sourcs.Objective:Combined with the research status of Rosa davurica Pall.,the separation andpurifaction of total flavonoids,the quality and HPLC fingerprint of the extracts hadbeen studied,which might provide a theoretical basis for the further application of thefruit.Methods:(1) D-101macroporous adsorption resin was employed to determine the optimalpurification process with single factor method.(2) Hyperoside in extracts of the fruit was qualitatively identified by thin layerchromatography;the content of total flavonoids, quercetin and gallic were determinedby UV and HPLC respectively.(3) The HPLC fingerprinting of the extracts of the fruit was established usingrutin,hyperin and quercetin as references.Results:(1) The content of the sample,optium loading quantity sample,adsorption time,pH value of sample,adsorption rate,ethanol content,desorption rate,the volume ofeluant were0.025g/mL,0.23g/g,30min,2to4,1-2BV/h,70%ethanol,1BV/h,and3times column volume respectively,which promote the purification of total flavonoidsto above80%.(2) In optimum TLC condition, the fluorescent spots of hyperin in correspondingsamples was clear.The optimum coloration condition of total flavonoids with UVmethod was obtained.The content of quercetin and gallic acid in extracts by HPLCmethod were0.22%and0.26%respectively.(3) Thirteen batches samples have shown high similarity under optimum HPLCcondition. Conclusions:(1) The experimental results have shown that purification effect with D-101macroporous adsorption resin is fit and easy to operate.(2) TLC method for the identification had a high specificity and methods todetermine the content was accurate and reliable.(3) The HPLC fingerprinting of the extracts of the fruit established in the papermight be applied for the quality control of the fruit extracts.
