The Fruit of Rosa davurica Pall.?FRDP?is the mature fruit of the mountain thorn rose,it is riched in flavonoids,polyphenols,saponins and other chemical components.Pharmacological experiments showed that the FRDP was a Chinese medicinal material that can be added to function foods.It had excellent anti-fatigue,anti-oxidation,and immunity-enhancing physiological activities.The optimal extraction process,purification process,quality control of the active ingredients in FRDP were explored,which may be used as raw materials in related function foods.First,the extraction process of total flavonoids and total polyphenols from FRDP were studied by single factor and orthogonal experiments.The results showed that the optimal extraction process of total flavonoids was as follows:the extraction solvent was 75%ethanol aqueous solution,and the ratio of material to liquid was 1:6?g:mL?.The mixture was heated and refluxed for 3 times with 5.0 heach time.The extraction efficiency of total flavonoids was 29.17mg/g and the average yield was 22.00%.The best extraction method of total polyphenols was ionic liquid synergistic ultrasonic-microwave assisted extraction method.The optimal extraction conditions were as follows:extraction solvent was 50%ethanol aqueous solution.The material to liquid ratio,water bath temperature and ultrasonic power were1:50?g:mL?,80°C and 200 W,respectively.The ultrasonic treatment,microwave power and microwave treatment were set 30min,200 W and 15 min with addition of 0.1 mol/L 1-butyl-3-methylimidazole and extending extraction for 1.5 h.The extraction efficiency of total polyphenols was 13.29 mg/g.The above research provided the basis for the extraction of the active ingredients of the FRDP.Secondly,the purification effect of macroporous resin method and metal complex method on total flavonoids of FRDP were investigated.The results showed that the metal complex method had a poor purification effect on the total flavonoids of FRDP,while the macroporous resin method was more suitable for the purification of FRDP.The results showed that the total flavonoids content in extract of FRDP could be improved rom about 25%to about 70%after purification by D-101 macroporous resin.Then the content of active ingredients in the fruit and its extracts was determined.The content of total flavonoids,total polyphenols,total saponins and tannins in extracts of FRDP were determined by UV-visible spectrophotometry.The content of hyperoside and luteolin in FRDP were determined by high performance liquid chromatography?HPLC?.According to the test results of various determination methods,the contents of total flavonoids,total polyphenols,total saponins and tannins in the extracts of FRDP were about 34%,41%,22%and 3%,respectively.The content of hyperoside and luteolin were 25.36?g/g and 11.99?g/g,respectively with HPLC.Furthermore,the preparation process of total flavonoids-Zn2+complex and its antioxidant activity in vitro were studied.Under the optimal preparation conditions,the yield of total flavonoids-Zn2+complex was 3.57%.The complex had certain scavenging ability to DPPH·,·OH,O2-·,·ABTS+,nitrite and so on.Finally,the preparation process of the total flavonoids capsules of FRDP function food was explored.The optimum preparation conditions of the capsules were determined by the particle forming rate and the disintegration time limit.The capsules were prepared under the conditions fit for the relevant requirements in Chinese Pharmacopoeia 2015 Edition.
