Preparation Of Dracocephalum Tanguticum Maxim. Drug Combination

Dracocephalum tanguticum Maxim. (DtM) drug combination is composed of DtM Astragalus, Codonopsis etc. It has good anti-hypoxia effect, can be used to prevent and relieve discomfort initiated by hypoxia. The combination is studied on preparation, pharmacology and toxicology in this subject.Based on properties of the prescription, the dosage form was chosen as capsule. On the basis of the function and the properties of the component included in the herbs, DtM was extracted by alcohol and others were extracted by water. Methods with total saponins contents and dry extract rate as the inspection indicators, the optimum alchohol-extract condions of DtM were that:the alcohol concentration was 60%, the solid-liquid ratio was 1:8, extraction time was 1 h, and the extraction times were 3 times. At the same time, methods with total polysaccharides contents and dry extract rate as the inspection indicators, the optimum water-extract condions of Codonopsis etc were that:the solid-liquid ratio was 1:8, extraction time was 0.5 h, and the extraction times were 3 times. Dose proportion of DtM and Codonopsis etc was determined as 10:1 by anti-hypoxia experiment under normal pressure. The forming process of capsule was as follows: weighted dry extract power in proportion, added edsuitable excipient to the extract power to make granulation, then made capsule.The results of effect experiment indicated that DtM drug combination can resist normobaric hypoxia and acute cerebral hypoxia-ischemia, especially the effect of anti-cerebral anoxia was signifcant. In addition, the combination had antioxidant capacity. It enhanced activity of glutathione peroxidase both in serum and liver and activity of glutathione in serum, decreased malondialdehyde content in liver. The results of acute toxicity test showed that LD50 of the combination more than 87.9 g/kg.The preliminary quality standard of the DtM drug combination had been made, indicating astragaloside which was component of the DtM drug combination can be used to qualitative identification.
