Trichloroethylene is a soluble substance which is used in industry .but it was poison to people's health. Aniline Is a important raw material substance in the industry.but it can cause animal and the mankind disease through many kinds of waysuch as suck it or touch it. .Phenol is the important raw material or midst substance of paper making, cokingplant, oil refining, plastic, pesticide and medicine composing etc. More and more phenol wastewater without treatment is threatening the environment heavily with the development of economy. It is economic and effective that using the method of microorganism biodegradation to treat with phenol wastewater. And it has not second pollution too.A strain that can grow with phenol as the sole source of carbon and energy, Y2(83), was isolated from the soil samplesin the GuangXi university wastewater at Nanning suburban. On the basis of observing of modality and the analysis of the 16SrDNA sequence, Y2[83] was identified as Mythylosistis primarily. About 242bp fragment which homologous to mmoX in sMMO gene cluster of Methylocuccos, was amplified by PCR, using conservative sequence of mmoX gene as primers. The fragment was sequenced and indicated that it has 85% homology to the mmoX gene of Methylocuccos which reported before. The radioactively labeled PCR product was used as probe to hybridize with the total DNA of Y2(83) which cut by EcoRI of restriction endonucleases, the result showed the sMMO gene's location. After constructed a sMMO gene's partial library, positive clone which including sMMO gene had been found by in situ hybridization at last. Our work laid a funcdation for reseaching sMMO gene.
