The similarities in the genomic organization, morphology, physical and chemical properties, molecular biological properties and pathogenicity between simian immunodeficiency virus (SIV) and human immunodeficiency virus (HIV) have made macaques with SIV/SHIV infection be used as an ideal model for various studies on AIDS. The SIV antigen detective assays are important to early diagnosing of primary viraemia, monitoring the efficacy of antiretroviral medicines and estimating disease progression. Among the structural proteins of SIV, p27 capsid protein (gag encoded) has been recognized as the surrogate marker of SIV antigen detection because of the conserved amino acid sequence and common antigenicity. In this thesis, the purified SIV p27 protein was used as antigen to immunize BALB/c mice and hybridoma cell lines that secreted anti-p27 antibodies were established. By using the produced monoclonal antibodies, we tried to develop the double-antibody sandwich ELISA method to detect SIV p27 antigen from free viruses in blood.Our studies included two parts. The studies in the first part were on making hybridoma cells for producing monoclonal antibodies (McAbs) against SIV p27 protein, on identifying and characterizing these McAbs. Two BALB/c mice were immunized with plasmid SIV p27 protein.
