Many areas of modern technology are dependent on sensitive analytical techniques, and this is particularly true for environmental science, medicine, pharmacy, and cellular biology. Labeling is one of the most common methodologies used for bioanalytical purposes. And rhodamine dyes are a family of 3,6-di (substituted amino)-9-benzoate derivatives of xanthene. Due to their excellent photophysical properties, such as long wavelength absorption and emission, high fluorescence quantum yield, large extinction coefficient, and high stability against light, they are widely used as fluorescent probes. A great interest in the development of new synthetic procedures for preparation of rhodamine derivatives has arisen in recent years.This paper describes the recent research of rhodamine fluorescent dyes. At first, the structure characteristics of them have been introduced. And then the synthesis of them is given in detail, according to the modification of different position of the rhodamine derivative moleculars. The strategies for synthesis of rhodamine derivatives and the relationship between the structure and function of the rhodamine molecular are summarized, highlightting the significance of their applications in the field of biomolecular labeling and environmental analysis.And then, the main results of our work about rhodamine probe are summarized as follows:1. The synthesis of 7-hydroxy-1,2-dihydroquinoline derivativesWe have found a method for producing 7-hydroxy-1,2-dihydroquinoline derivatives which is not only simple and efficient but which also results the desired product utilizing readily available and inexpensive starting materials.2. The synthesis of rigid rhodamine dyesA series of novel rigid rhodamine dyes were synthesized by condensation of certain 7-hydroxy-1,2-dihydroquinoline derivatives with phthalic anhydride. A novel method for preparation of unsymmetrical rhodamine dyes had been tried out. The obtained dyes have long wavelength absorption and emission.3. The synthesis of rhodamine fluorescent probe Individual isomers of 4'-and 5'-carboxy-X-rhodamines have been successfully synthesized and separated. A novel kind of rhodamine-based fluorescent probe dye with spacer linker arm was designed and synthesized.
