Naturalβ-carotene has many healthful function,such as anti-cancer, improve immunity and delay caducity health. The development of extraction and new products about Naturalβ-carotene has been increasing attention in the field of health and medical applications. In order to improve value-added of carrots and research new products, we studied conditions of microwave-assisted method extraction onβ-carotene yield, explored the process parameters of naturalβ-carotene ,further made a systematic study on stability and biological activity of products;The results were as follows:.(1) Named zhong jia yi hao variety of carrot grown in Xinjiang as raw material,β-carotene content was up to 252.20mg/100g fresh carrot, The material were ground by a mill to pass a 80-mesh size screen ; In order to absorb microwave energy effectively and improve extracting rate,It was better to impregnate material with 75% ethanol before extraction.(2) The tests were optimized through good single factor selecting and Response Surface Methodology,studied on microwave-assisted extraction ofβ-carotene in this test.The predicted values were obtained by equation very well.To ethyl acetate as solvent, the extraction optimal conditions is microwave power 386W, time 7min and rate of liquid /solid 22,β- carotene extraction reached 0.5587mg / g; In soybean salad oil as the solvent, the optimal conditions is microwave power 394W, time 6min, and rate of liquid / solid 43,β-carotene extraction amounted to 0.399mg / g. Application of ethyl acetate in the extraction from carrot power heads dramatically reduced extraction rate of liquid /solid and significantly improve the naturalβ-carotene extraction yield compared with soybean salad oil.(3) Study on the process of naturalβ-carotene soft capsule, and the optimum process parameters: melt gelatin temperature60-70℃, melt gelatin time of 4 hours, vacuum degree-0.08-0.06Mpa; pressed pill process parameters: spray temperature 38-45℃, filling temperature<40℃, gelatin liquid temperature 55-65℃.The soft capsule was mesurated about micro-organisms, heavy metals, disintegration time,acid value,the net and so on,the result was according with healthy food request.(4) The study of the capsule under the test of influencing factors demonstrated that the capsule was stable at high temperature,high humidity and sun;The stability experiment of naturalβ-Carotene soft capsule under normal conditions,the result was showed: Microbe index,physical and chemical index and Sense identification. sense index such as tissue sate did not have significant change in three months,The index comply to healthy food request published by national sanitation ministry。(5) In order to discuss natural naturalβ-carotene soft capsule antioxidant activities, adopted NO2-,.OH,O2-and DPPH free radical scavenging capacity.The result indicated that the O2- elimination ability of the soft capsule was strong, followed by NO2- and DPPH free radicals and the weakest scavenging was.OH. Compared to VC, at the selected concentration range, the soft capsule and VC on the NO2- elimination ability was equally effect, the effect of O2- scavenging was less than VC's, but at concentrations lower than 54ug/mL ,the the soft capsule elimination ability is stronger than VC's for.OH.(6) By MTT and microscopic observation,the inhibitory effect of naturalβ-Carotene soft capsule on the growth and proliferation of S180 cells was was not significant at the selected concentration range,the inhibition mechanism is to be studied further.
