Honey is a natural health food, the research on antoxidative activity and prevention effects to oxidative DNA damage of honey has a significant meaning for investigating its health protection functions deeply. The antoxidative activity and prevention effects to oxidative DNA damage of 8 batches honey samples were studies in this paper. A novel flow injection chemiluminescence method was established for antoxidative ability and DNA oxidative damage protection capabilities of all honey samples, and this method was applied in evaluation the oxidative DNA damage protection capabilities of Buckwheat honey, Longan honey, Acacia honey and Rape honey. Moreover, its protective mechanism was also studied. The full text is divided into four chapters, the main contents are as follows:1, Review the progress of DNA oxidative damage.2, The antoxidative ability of 8 batches honey were measured using FI-CL method. Namely, the superoxide anion scavenging ability, hydroxyl radical scavenging ability and hydrogen peroxide scavenging ability. The results show that Buckwheat honey has the strongest ability in ROS scavenging based on the Comparison of the antoxidative capabilities of all honey samples.3, A novel flow injection chemiluminescence method was established based on the CUSO4-H2O2-DNA system. And this method was explored to evaluate the DNA oxidative damage protection capabilities of all honey samples. Then, the mechanism of CUSO4-H2O2-DNA system was investigated. The experimental results show that a chemiluminescence phenomenon was observed in the process of interaction between DNA and Fenton like reagent including Cu2+ and H2O2, the signal of the system was decreased sharply when honey sample added into system. This because honey has a strong ability of protect against DNA damage, the experiment indicates that Buckwheat honey has a strongest ability in protect against DNA damage among all honey samples. This ability may own to the highest concentration of TPC.4, The Agarose gel electrophoresis, ELISA, AFM, CD and SWV were used in the experiment about study the mechanism oxidative DNA damage and the protection effect of honey. The results shows that H2O2 could produce the hydroxyl radical when Cu2+ act as Catalyst. The hydroxyl radical will cut the double helix structure of DNA to form some DNA fragments, which induces a conformational change to DNA and oxidative damage products such as 8-OHdG were generated in the system. A certain influence on the structure of DNA when honey was added. Therefore, honey could protect DNA from oxidative damage. The substations such as phenolic acid in honey could complex the ions to reduce the generation of hydroxyl radicals.
